Thus, ‘digital constitutionalism’ emerged as a twin discourse. A corollary of these discourses is a third phenomenon resulting from a regulatory externalisation of European law beyond the bloc’s borders, the so-called ‘Brussels Effect’. The dynamics arising from Europe’s digital policy and regulatory activism imply increasing legal complexities. This paper argues that this phenomenon in policy-making is a case of a positive ‘policy bubble’ characterised by an oversupply of policies and legislative acts. The phenomenon can be explained by the amplification of values in the framing of digital policy issues. To unpack the policy frames and values at stake, this paper provides an overview of the digital policy landscape, followed by a critical assessment to showcase the practical implications of positive policy bubbles. The MoS2 layer is modeled as a geometrically nonlinear Kirchhoff–Love shell, and vdW forces are modeled by a Lennard-Jones (LJ) potential, simplified using approximations for a smooth substrate topography. Both the shell model and LJ interactions include novel extensions informed by close comparison with fully-atomistic calculations. The material parameters of the shell model are calibrated by comparing small-strain tensile and bending tests with atomistic simulations. This model is efficiently discretized using isogeometric analysis (IGA) for the shell structure and a pseudo-time continuation method for energy minimization. The IGA shell model is validated against fully-atomistic calculations for several benchmark problems with different substrate geometries. Agreement with atomistic results depends on geometric nonlinearity in some cases, but a simple isotropic St.Venant–Kirchhoff model is found to be sufficient to represent material behavior. We find that the IGA discretization of the continuum model has a much lower computational cost than atomistic simulations, and expect that it will enable efficient design space exploration in strain engineering applications. This is demonstrated by studying the dependence of strain and curvature in MoS2 over a holey substrate as a function of the hole spacing on scales inaccessible to atomistic calculations. The prediction of the time-dependent response of such media requires consideration of their characteristic multi-scale and multi-physics parameters. In the present work this is achieved by formulating a non-dimensionalised fluid–solid interaction problem (FSI) at the pore level using an arbitrary Lagrange–Euler description (ALE). The resulting coupled systems of PDEs on the reference configuration are expanded and analysed using the asymptotic homogenisation technique. This approach yields three partially novel systems of PDEs: the macroscopic/effective problem and two supplementary microscale problems (fluid and solid). The latter two provide the microscopic response fields whose average value is required in real-time/online form to determine the macroscale response (a concurrent multi-scale approach). In order to overcome the computational challenges related to the above multi-scale closure, this work introduces a surrogate approach for replacing the direct numerical simulation with an artificial neural network. This methodology allows for solving finite strain (multi-scale) porohyperelastic problems accurately using direct automated differentiation through the strain energy. Optimal and reliable training data sets are produced from direct numerical simulations of the fully-resolved problem by including a simple real-time output density check for adaptive sampling step refinement. The data-driven approach is complemented by a sensitivity analysis of the RVE response. The significance of the presented approach for finite strain poro-elasticity/poro-hyperelasticity is shown in the numerical benchmark of a multi-scale confined consolidation problem. Finally, to show the robustness of the method, the system response is dimensionalised using characteristic values of soil and brain mechanics scenarios. In this study, we investigated the lattice dynamics of PbVO3 based on Raman spectroscopy at room temperature and first-principle calculations. All zone-center transverse optical phonon modes were identified by polarized, angle-dependent Raman spectroscopy and assigned as follows: E modes at 136, 269, 374, and 508 cm−1; A1 modes at 188, 429, and 874 cm−1, and B1 mode at 319 cm−1. The calculations confirmed the experimental symmetry assignment and allowed us to obtain the longitudinal optical phonon wavenumbers. In addition, we analyzed the mode eigenvectors in detail in order to identify the atomic displacements associated with each mode and compare them with PbTiO3. Despite differences in chemistry and strain, the phonon eigenvectors were found to be highly comparable in both compounds. We investigated the position of the ferroelectric soft mode in PbVO3 compared with PbTiO3. Sizeable splitting of the B1+E modes appeared as a characteristic feature of supertetragonal phases. Starting with an existing published data set on melt instabilities in polymer extrusion, we assess the suitability of clustering, unsupervised machine learning algorithms combined with feature selection, to extract and identify hidden and important features from this data set, and their possible relationship with melt instabilities. The data set consists of both intrinsic features of the polymer as well as extrinsic features controlled and measured during an extrusion experiment. Using a range of commonly available clustering algorithms, it is demonstrated that the features related to only the intrinsic properties of the data set can be reliably divided into two clusters, and that in turn, these two clusters may be associated with either the stick-slip or sharkskin instability. Furthermore, using a feature ranking on both the intrinsic and extrinsic features of the data set, it is shown that the intrinsic properties of molecular weight and polydispersity are the strongest indicators of clustering. In particular, 4 cars have been connected to this platform for periods of over one month. Our results highlight that, while this technological innovation appears very promising in the future, the pricing, the lack of uniformity of data collected and the enrollment process are currently three pain points that should be addressed to offer large-scale opportunities. We utilized mass spectrometry to investigate the proteome of CD14(+) monocytes in healthy adults reporting childhood adversity and a control group before and after psychosocial stress exposure. Particular proteins involved in (i) immune processes, such as neutrophil-related proteins, (ii) protein metabolism, or (iii) proteins related to mitochondrial biology, such as those involved in energy production processes, were upregulated in participants reporting exposure to adversity in childhood. This functional triad was further corroborated by protein interaction- and co-expression analyses, was independent of stress exposure, i.e. observed at both pre- and post-stress time points, and became evident especially in females. In line with the mitochondrial allostatic load model, our findings provide evidence for the long-term effects of childhood adversity on mitochondrial biology. The analysis shows evidence of brain drain in Taiwan since the 2010s, with the U.S. being both the biggest source of inbound researchers to Taiwan and top destination for researchers emigrating from Taiwan. China comes a close second to the U.S. as the top destination for outbound scholars. We also study how Taiwan’s universities recruited talent after the country adopted the 2005 excellence initiative and confirm the converging number of scholars recruited by World Class Universities (WCUs) and non-WCUs as WCUs, surprisingly, exhibit a dramatic decrease in new recruits. Our evidence uncovers that inbound scholars, after their move, are more productive than non-mobile colleagues; however, this effect declines over time. We discuss implications for the study of excellence initiatives and mechanisms of talent circulation that greatly impact research production and research university development. In the recent few years, establishing multi-layer NTNs has been extensively studied to integrate space-airborne-terrestrial communications systems, particularly by the international standardization organizations such as the third-generation partnership project (3GPP) and the international telecommunication union (ITU), in order to support future wireless ecosystems. Indeed, amalgamating quantum technologies and NTNs will scale up the quantum communications ranges and provide unprecedented levels of security and processing solutions that are safer and faster than the traditional offerings. This paper provides some insights into the interplay between the evolving NTN architectures and quantum technologies with a particular focus on the integration challenges and their potential solutions for enhancing the quantum-NTN interoperability among various space-air-ground communications nodes. Yet, so far, we lack knowledge about the relationship between economic inequality and the prevalence of family firms in an economy. Our study investigates how the share of family-controlled public firms correlates with various measures of income and wealth inequality. The results show that a higher share of public family-controlled firms leads to more income inequality in a country. This effect is particularly pronounced for the middle of the income distribution as opposed to the top quantiles. Redistribution only mitigates this effect to some extent, as the effect is significant for market income and disposable income. We also find that a higher share of family-controlled firms contributes to an increase in wealth inequality. Our results are of economic relevance as, for instance, a one standard deviation change in the share of family-controlled firms leads to an increase of around 1.3–1.5 percentage points in the Gini coefficients for market income, disposable income, and wealth. Delivery methods could broadly be classified into group classes, individual support and peer learning. Microteaching with reflection, feedback and observation was a widely recurring and highly rated activity. Programmes were typically developed in-house by English language professionals. Recurring challenges were contextualisation, group heterogeneity, lecturer confidence and the lack of incentivisation. They are typically solved using Lagrangian-Eulerian methods, one of which is the coupling between the discrete element method (DEM, which is applied to the particles phase) and the computational fluid dynamics method (CFD, which is applied to the fluid phase). The current study's objective is to avoid coupling and instead, focusing on using the CFD method only. There are dense piles of particles moving on the grates in the biomass combustion chamber. We assumed the dense particles' behaviors similar to the fluid, and then, applied the fluid governing equations to the particles phase. The virtual fields of the velocities, pressure and density are specified for the particles' phase. Afterward, the physics-informed neural network (PINN) is used to reconstruct particles' fields and additionally to investigate the capability of the predicted fields to satisfy the fluid governing equations. This model has the benefit of reconstructing the particles' fields without the need for boundaries and initial conditions. The precision of the model is assessed by comparing the test data set with the exact data obtained from the eXtended discrete element method (XDEM is an in-house software). It is demonstrated that the trained neural network delivered high accuracy and is capable of predicting all outputs with an error value of less than 2 percent. Additionally, to choose the optimum architecture for the neural network, the effect of the number of hidden layers and neurons is studied. The current study seeks to investigate kindergarten children’s attitudes towards homosexuality and how they are influenced by their peer-groups and authorities. It does so by means of a puppet theatre presentation including ‘Kasperl’ and ‘Seppel’ puppets as the main protagonists in a German puppet theatre. The audience consisted of six German kindergarten children aged 4–6 years, randomly selected by the kindergarten director. A participant observation has been conducted with the first researcher as the presenter of the puppet show. The results of the recorded puppet theatre showed that several intragroup processes including the social identity theory with regard to group membership could be observed within the puppet theatre. Groups have been formed and children of the same ingroup showed equal attitudes towards homosexuality. However, the researcher’s influence as an authority figure could have as well impacted their attitudes through positive reinforcement. The present study contributed to the gap in research regarding kindergarten children’s attitudes towards homosexuality. To have a better overview of the results, future studies should include additional variables like kindergarten’s cultural and geographical differences. The final sample consisted of N = 72 fifteen-month-old infants and their mothers. Mothers completed a survey on pre- and postnatal negative life events. To assess chronic stress physiologically, infant and maternal hair cortisol concentrations were determined for cortisol accumulation during the past 3 months. Each infant participated in two cognitive tasks in the laboratory. An instrumental learning task tested infants’ ability to disengage from a habituated action when this action became ineffective (Seehagen et al., 2015). An age-adequate version of the A-not-B task tested infants’ ability to find a toy at location B after repeatedly finding it at location A. Correlations between cortisol concentrations and postnatal negative life events (number, perceived impact) did not yield significance. Infant and maternal hair cortisol concentrations were not correlated. Infants’ ability to shift to a new action in either task, controlled for acute stress, correlated neither with pre- and postnatal negative life events nor with cortisol concentrations. Taken together, these results indicate that the potential link between long-term stress exposure and cognitive flexibility might not be present in samples with low levels of psychosocial stress. The suppressive functions of Ambra1 in melanoma are mediated by negative regulation of cell proliferation and invasion; however, evidence suggests that loss of Ambra1 may also affect the melanoma microenvironment. Here, we investigate the possible impact of Ambra1 on antitumor immunity and response to immunotherapy. METHODS: This study was performed using an Ambra1-depleted Braf(V600E) /Pten(-/) (-) genetically engineered mouse (GEM) model of melanoma, as well as GEM-derived allografts of Braf(V600E) /Pten(-/) (-) and Braf(V600E) /Pten(-/) (-)/Cdkn2a(-/) (-) tumors with Ambra1 knockdown. The effects of Ambra1 loss on the tumor immune microenvironment (TIME) were analyzed using NanoString technology, multiplex immunohistochemistry, and flow cytometry. Transcriptome and CIBERSORT digital cytometry analyses of murine melanoma samples and human melanoma patients (The Cancer Genome Atlas) were applied to determine the immune cell populations in null or low-expressing AMBRA1 melanoma. The contribution of Ambra1 on T-cell migration was evaluated using a cytokine array and flow cytometry. Tumor growth kinetics and overall survival analysis in Braf(V600E) /Pten(-/) (-)/Cdkn2a(-/) (-) mice with Ambra1 knockdown were evaluated prior to and after administration of a programmed cell death protein-1 (PD-1) inhibitor. RESULTS: Loss of Ambra1 was associated with altered expression of a wide range of cytokines and chemokines as well as decreased infiltration of tumors by regulatory T cells, a subpopulation of T cells with potent immune-suppressive properties. These changes in TIME composition were associated with the autophagic function of Ambra1. In the Braf(V600E) /Pten(-/) (-)/Cdkn2a(-/) (-) model inherently resistant to immune checkpoint blockade, knockdown of Ambra1 led to accelerated tumor growth and reduced overall survival, but at the same time conferred sensitivity to anti-PD-1 treatment. CONCLUSIONS: This study shows that loss of Ambra1 affects the TIME and the antitumor immune response in melanoma, highlighting new functions of Ambra1 in the regulation of melanoma biology. To date, however, it is not clear whether and based on which molecular mechanisms microglia contribute to the development and progression of focal epilepsy. Astroglial gap junction coupled networks play an important role in regulating neuronal activity and loss of interastrocytic coupling causally contributes to TLE. Here, we show in the unilateral intracortical kainate (KA) mouse model of TLE that reactive microglia are primary producers of tumor necrosis factor (TNF)α and contribute to astrocyte dysfunction and severity of status epilepticus (SE). Immunohistochemical analyses revealed pronounced and persistent microglia reactivity, which already started 4 h after KA-induced SE. Partial depletion of microglia using a colony stimulating factor 1 receptor inhibitor prevented early astrocyte uncoupling and attenuated the severity of SE, but increased the mortality of epileptic mice following surgery. Using microglia-specific inducible TNFα knockout mice we identified microglia as the major source of TNFα during early epileptogenesis. Importantly, microglia-specific TNFα knockout prevented SE-induced gap junction uncoupling in astrocytes. Continuous telemetric EEG recordings revealed that during the first 4 weeks after SE induction, microglial TNFα did not significantly contribute to spontaneous generalized seizure activity. Moreover, the absence of microglial TNFα did not affect the development of hippocampal sclerosis but attenuated gliosis. Taken together, these data implicate reactive microglia in astrocyte dysfunction and network hyperexcitability after an epileptogenic insult. Cancer-associated fibroblast (CAF) heterogeneity is characteristic of the pancreatic TME, where several CAF subpopulations have been identified, such as myofibroblastic CAFs (myCAFs), inflammatory CAFs (iCAFs), and antigen presenting CAFs (apCAFs). In PDAC, cancer cells continuously adapt their metabolism (metabolic switch) to environmental changes in pH, oxygenation, and nutrient availability. Recent advances show that these environmental alterations are all heavily driven by stromal CAFs. CAFs and cancer cells exchange cytokines and metabolites, engaging in a tight bidirectional crosstalk, which promotes tumor aggressiveness and allows constant adaptation to external stress, such as chemotherapy. In this review, we summarize CAF diversity and CAF-mediated metabolic rewiring, in a PDAC-specific context. First, we recapitulate the most recently identified CAF subtypes, focusing on the cell of origin, activation mechanism, species-dependent markers, and functions. Next, we describe in detail the metabolic crosstalk between CAFs and tumor cells. Additionally, we elucidate how CAF-driven paracrine signaling, desmoplasia, and acidosis orchestrate cancer cell metabolism. Finally, we highlight how the CAF/cancer cell crosstalk could pave the way for new therapeutic strategies. This raises the question of whether the task would be a useful component of mathematical achievement tests and instruments to diagnose dyscalculia or mathematical giftedness and whether a stand-alone version of the task can serve as a short screener for mathematical achievement. Previous studies on the relation between number line estimation accuracy and broader mathematical achievement were limited in that they used relatively small nonrepresentative samples and usually did not account for potentially confounding variables. To close this research gap, we report findings from a population-level study with nearly all Luxembourgish ninth-graders (N = 6484). We used multilevel regressions to test how a standardized mathematical achievement test relates to the accuracy in number line estimation on bounded number lines with whole numbers and fractions. We also investigated how these relations were moderated by classroom characteristics, person characteristics, and trial characteristics. Mathematical achievement and number line estimation accuracy were associated even after controlling for potentially confounding variables. Subpopulations of students showed meaningful differences in estimation accuracy, which can serve as benchmarks in future studies. Compared with the number line estimation task with whole numbers, the number line estimation task with fractions was more strongly related to mathematical achievement in students across the entire mathematical achievement spectrum. These results show that the number line estimation task is a valid and useful tool for diagnosing and monitoring mathematical achievement. In order to characterize such communication blackout phases, this work presents a numerical methodology combining Computational Fluid Dynamic (CFD) simulations of ionized chemically reacting entry flows by means of Computational Object-Oriented Libraries for Fluid Dynamics (COOLFluiD) and a ray tracing analysis by means of the newly developed BlackOut RAy Tracer (BORAT). The latter is based on the numerical solution of the 3D Eikonal system of equations, offering a fast, efficient and accurate method to analyse the interaction between electromagnetic signals and weakly ionised plasmas. The proposed methodology, and BORAT in particular, is first verified on popular benchmark cases and then used to analyse the European Space Agency (ESA) 2016 ExoMars Schiaparelli entry flight into Martian environment. The corresponding results demonstrate the validity of the proposed ray tracing approach for predicting communication blackout, where signals emitted from the on-board antenna undergo reflection and refraction from the plasma surrounding the entry vehicle, and the advantage of a 3D approach for analysing real flight configuration. Subsequently, it was examined to which extent gender differences on these tests could be explained by verbal and visuo-spatial working memory, explicit and implicit gender-related stereotypes and math anxiety. Results showed that women scored significantly lower than men on the arithmetic and cognitive reflection tests. A mediation analysis demonstrated that the relation between gender and arithmetic performance was partially mediated by math anxiety and explicit gender-related stereotypes. Furthermore, results showed that math anxiety fully mediated the relation between gender and cognitive reflection. These results demonstrate that math anxiety plays a key role in the relation between gender and mathematical performance. Since GBM is a rare disease, sufficient statistically strong evidence is often not available to explore the roles of lesser-known GBM proteins. We present a network-based approach using centrality measures to explore some key, topologically strategic proteins for the analysis of GBM. Since network-based analyses are sensitive to changes in network topology, we analysed nine different GBM networks, and show that small but well-curated networks consistently highlight a set of proteins, indicating their likely involvement in the disease. We propose 18 novel candidates which, based on differential expression, mutation analysis, and survival analysis, indicate that they may play a role in GBM progression. These should be investigated further for their functional roles in GBM, their clinical prognostic relevance, and their potential as therapeutic targets. Existing methods are generally trained in a supervised manner at the classification level, focusing on detecting at most two types of forgeries; thus, limiting their generalization capability across different deepfake types. To handle that, we reformulate the deepfake detection problem as a one-class classification supported by a self-supervision mechanism. Our intuition is that by estimating the distribution of real data in a discriminative feature space, deepfakes can be detected as outliers regardless of their type. UNTAG involves two sequential steps. First, deep representations are learned based on a self-supervised pretext task focusing on manipulated regions. Second, a one-class classifier fitted on authentic image embeddings is used to detect deepfakes. The results reported on several datasets show the effectiveness of UNTAG and the relevance of the proposed new paradigm. The code is publicly available. First results indicate a mediating role of interoception in the interplay of traumatic childhood experiences and adult mental disorders. Traumatic childhood experiences may hinder the adequate processing, integration, and trust in bodily signals that are important in order to understand and regulate own needs and emotions, thereby increasing the vulnerability for mental disorders. However, an overarching study investigating alterations in different interoceptive measures and trauma-related disorders as well as their mediating role between early trauma and emotion dysregulation is still missing. Methods One hundred thirty-six individuals with varying levels of traumatic childhood experiences who either had a current diagnosis of major depression, posttraumatic stress disorder, or somatic symptom disorder, or no mental disorder, took part in a multidimensional assessment of interoceptive processes, including interoceptive accuracy, sensibility, and awareness. Kruskal–Wallis tests were used to compare groups regarding interoceptive processes and associations with traumatic childhood experiences and emotion dysregulation were analyzed with Spearman correlations. Furthermore, mediation analyses were computed to examine and compare interoceptive processes as potential mediators between traumatic childhood experiences and emotion dysregulation. Results Only body dissociation, a measure for interoceptive sensibility, was significantly reduced in individuals with a current mental disorder. Body dissociation was also the only interoceptive measure significantly associated with traumatic childhood experiences and emotion dysregulation and the only significant mediator in the relationship between traumatic childhood experiences and emotion dysregulation across groups. Conclusion Results suggest body dissociation, but not other interoceptive measures, as an important feature linking traumatic childhood experiences to current emotion dysregulation, an important transdiagnostic feature. As body dissociation refers to a habitual non-attendance or disregard of interoceptive signals, integrative therapeutic interventions could help affected individuals to overcome difficulties in emotion perception and regulation. A wire rope consists of numerous thin, steel wires and its geometrical construction can be explained in two steps. First, several wires are wrapped together in a helical shape called a strand. Second, several strands are wrapped together in a helical shape to form the final wire rope. In most cases, each strand is compacted before they are wrapped together to form the final wire rope. Compaction generally reduces contact stresses and thereby, extends ropes’ service life. Not many models have been proposed to predict the compaction process and its influence on the strand’s mechanical behavior during service. This contribution proposes a computationally efficient approach that consists of two elastoplastic mechanical models. The first model, describing the compaction process, is of a 2D plane strain nature and is therefore fast. Subsequently, the 2D geometry and plastic variables predicted by the compaction model are used to generate the initial geometry and initial plastic state of a 3D model, that is subsequently used to describe the strand’s mechanical behavior during service (we limit ourselves to tension). The results of the approach, with and without the mapping of the plastic variables, are compared to experimental measurements and the results without compaction. The machine learning approach was trained using data generated by four laboratories equipped with different instrumentation. The model discarded substances with insufficient identification evidence efficiently, while revealing the relevance of different parameters for identification. Based on these results, a harmonized IP-based system is proposed. This new NTS-oriented system is compatible with the currently widely used five level system. It increases the precision in reporting and the reproducibility of current approaches via the inclusion of evidence scores, while being suitable for automation. We propose here a test to find spliceQTL effects which takes all exons and all SNPs into account simultaneously. For any chosen gene, this score-based test looks for association between the set of exon expressions and the set of SNPs, via a random-effects model framework. It is efficient to compute, and can be used if the number of SNPs is larger than the number of samples. In addition, the test is powerful to detect effects that are relatively small for individual exon-SNP pairs, but are observed for many pairs. Furthermore, test results are more often replicated across datasets than pairwise testing results. This partly our test is more robust to exon-SNP pair-specific effects, but do not extend to multiple pairs within the same gene. Today, there exists a plethora of DLT platforms on the market, which makes it challenging for system designers to decide what platform they should adopt and implement. Although a few DLT comparison frameworks have been proposed in the literature, they often fail in covering all performance and functional aspects, adding that they too rarely build upon standardized criteria and recommendations. Given this state of affairs, the present paper considers a recent and exhaustive set of assessment criteria recommended by the ITU (International Telecommunication Union). Those criteria (about fifty) are nonetheless mostly defined in a textual form, which may pose interpretation problems during the implementation process. To avoid this, a systematic literature review regarding each ITU criterion is conducted with a twofold objective: (i) to understand to what extent a given criterion is considered/evaluated by the literature; (ii) to come up with ‘formal’ metric definition (i.e., on a mathematical or experimental ground) based, whenever possible, on the current literature. The validity of these approximations is demonstrated using extensive Monte Carlo simulations. We confirm the achievable rate improvement decided by main parameters such as the average signal to noise ratio at source, the position of IRS with respect to the source and destination and the number of IRS elements. As a suggestion for the deployment of a low-cost IoT system, the double-IRS model is a reliable approach to realizing the system as long as the hardware impairment level is controlled. Here, we present a computationally driven analytical review of epilepsy-associated genes using the complete scientific literature in PubMed. Based on our search criteria, we identified a total of 738 epilepsy-associated genes. We further classified these genes into two Tiers. A broad gene list of 738 epilepsy-associated genes (Tier 2) and a narrow gene list composed of 143 epilepsy-associated genes (Tier 1). Our search criteria do not reflect the degree of association. The average yearly number of identified epilepsy-associated genes between 1992 and 2021 was 4.8. However, most of these genes were only identified in the last decade (2010–2019). Ion channels represent the largest class of epilepsy-associated genes. For many of these, both gain- and loss-of-function effects have been associated with epilepsy in recent years. We identify 28 genes frequently reported with heterogenous variant effects which should be considered for variant interpretation. To overcome these challenges, an algorithm to detect homologous series within compound datasets was developed and implemented using the RDKit. The algorithm takes a list of molecules as SMILES strings and a monomer (i.e., repeating unit) encoded as SMARTS as its main inputs. In an iterative process, substructure matching of repeating units, molecule fragmentation, and core detection lead to homologous series classification through grouping of identical cores. Three open compound datasets from environmental chemistry (NORMAN Suspect List Exchange, NORMAN-SLE), exposomics (PubChemLite for Exposomics), and natural products (the COlleCtion of Open NatUral producTs, COCONUT) were subject to homologous series classification using the algorithm. One extensively studied components of liquid biopsies are exosomes, a subtype of extracellular vesicle. Due to their unique molecular cargo, they could be used as carriers of early markers of cancer development and progression. For characterisation of the miRNA profiles present in circulating serum-derived exosomes of patients with diagnosed primary and metastatic UM, we have analysed the miRNA cargos using next-generation sequencing followed by RT-qPCR validation in a cohort of patients (control n=20; primary n=9; metastatic n=11). Nine miRNAs clearly differentiating these patient groups have been established. We show that hsa-miR-223 and hsa-miR-203a are the most promising biomarker candidates, allowing categorization of patients into local and advanced UM. Additionally, the comparison of miRNA expression levels in exosomes derived from UM patients with those derived from healthy donors, revealed that hsa-miR-144 has the potential to be used as an early marker for presence of UM. Taken together, this pilot study reveals that miRNAs extracted from circulating exosomes could be exploited as potential biomarkers in UM diagnosis and, more importantly, for indicating metastatic spread. We find that the ordinary differential equations, which determine bound soliton solutions, can be transformed into the form resembling the Schrödinger-like equations for eigenfunctions with fixed eigenvalues. We assume that a nonlinear part of the equations is close to the reflectionless potential well occurring in the scattering problem associated with the integrable equations. Given the established role of PINK1/Parkin in regulating mitochondrial dynamics, we explored mitochondrial DNA (mtDNA) integrity and inflammation as disease modifiers in carriers of mutations in these genes. MtDNA integrity was investigated in a large collection of biallelic (n = 84) and monoallelic (n = 170) carriers of PINK1/PRKN mutations, idiopathic Parkinson’s disease patients (n = 67) and controls (n = 90). In addition, we studied global gene expression and serum cytokine levels in a subset. Affected and unaffected PINK1/PRKN monoallelic mutation carriers can be distinguished by heteroplasmic mtDNA variant load (AUC = 0.83, CI:0.74-0.93). Biallelic PINK1/PRKN mutation carriers harbor more heteroplasmic mtDNA variants in blood (p = 0.0006, Z = 3.63) compared to monoallelic mutation carriers. This enrichment was confirmed in iPSC-derived (controls, n = 3; biallelic PRKN mutation carriers, n = 4) and postmortem (control, n = 1; biallelic PRKN mutation carrier, n = 1) midbrain neurons. [more ▼] The digital transformation of the medical sector requires solutions that are convenient and efficient for all stakeholders while protecting patients’ sensitive data. One example that has already attracted design-oriented research are medical prescriptions. However, current implementations of electronic prescription management systems typically create centralized data silos, leaving user data vulnerable to cybersecurity incidents and impeding interoperability. Research has also proposed decentralized solutions based on blockchain technology, but privacy-related challenges have often been ignored. We conduct design science research to develop and implement a system for the exchange of electronic prescriptions that builds on two blockchains and a digital wallet app. Our solution combines the bilateral, verifiable, and privacy-focused exchange of information between doctors, patients, and pharmacies through verifiable credentials with a token-based, anonymized double-spending check. Our qualitative and quantitative evaluations as well as a security analysis suggest that this architecture can improve existing approaches to electronic prescription management by offering patients control over their data by design, a high level of security, sufficient performance and scalability, and interoperability with emerging digital identity management solutions for users, businesses, and institutions. The relevance of surface curvature in biology has been supported by numerous recent experimental and theoretical investigations in recent years. In this review, we first give a brief introduction to the key ideas of surface curvature in the context of biological systems and discuss the challenges that arise when measuring surface curvature. Giving an overview of the emergence of curvature in biological systems, its significance at different length scales becomes apparent. On the other hand, summarizing current findings also shows that both single cells and entire cell sheets, tissues or organisms respond to curvature by modulating their shape and their migration behavior. Finally, we address the interplay between the distribution of morphogens or micro-organisms and the emergence of curvature across length scales with examples demonstrating these key mechanistic principles of morphogenesis. Overall, this review highlights that curved interfaces are not merely a passive by-product of the chemical, biological and mechanical processes but that curvature acts also as a signal that co-determines these processes. While overall response speed was significantly faster when a deadline extension was communicated, the overall response rate was slightly lower, compared to scenarios in which no deadline extension was communicated. Thus, we refrain from recommending a deadline extension unless there are specific reasons to do so. Further, a low-complexity, suboptimal, yet efficient sensor source selection scheme is proposed. Specifically, one sensor source is chosen to transmit information to the relay and destination such that it obtains the best channel from sensor sources to the relay. Then, by considering two relaying strategies, termed the direct link plus static power splitting-based relaying (SPSR) and direct link plus optimal dynamic power splitting-based relaying (ODPSR), the performance analysis in terms of intercept probability (IP) and outage probability (OP) are carried out for each one. Notably, the eavesdropper and destination utilize maximal ratio combining (MRC) to incorporate the received signals from the selected sensor source and the relay, which poses new challenges in obtaining the analytical expressions. In this context, we derive analytical expressions for the OP (for SPSR and ODPSR) at the destination and the IP (for SPSR) at the eavesdropper by adopting the series representation of the modified Bessel function. Finally, Monte Carlo simulations are conducted to validate the theoretical analysis and the proposed schemes’ effectiveness. To make the best use of NOMA as an enabler for cell-free massive MIMO systems, user pairing should be employed effectively. Random user pairing naturally leads to a non-optimal solution, whereas an exhaustive search approach is unfavorable for practical systems owing to the high complexity. In this study, we propose an optimal user pairing strategy to group users that jointly optimize the minimum downlink rate per user and power allocation at an acceptable cost of complexity. To address this problem, we first relax the binary variables to continuous variables and then develop an iterative algorithm based on the inner approximation method, yielding at least one locally optimal solution. Numerical results show that the proposed user pairing algorithm outperforms existing counterparts, such as conventional beamforming, random pairing, far pairing, and close-pairing strategies, while it can be performed dynamically, that is, two arbitrary users satisfying the formulated problem can be paired regardless of geographical distance. Finally, our approach demonstrates that the combination channel estimation-based NOMA-assisted cell-free massive MIMO achieves the best result in terms of the downlink rate per user when associated with the proposed algorithm.
