NADHX and NADPHX are hydrated and redox inactive forms of the NADH and NADPH cofactors, known to inhibit several dehydrogenases in vitro. A metabolite repair system that is conserved in all domains of life and that comprises the two enzymes NAD(P)HX dehydratase and NAD(P)HX epimerase, allows reconversion of both the S- and R-epimers of NADHX and NADPHX to the normal cofactors. An inherited deficiency in this system has recently been shown to cause severe neurometabolic disease in children. Although evidence for the presence of NAD(P)HX has been obtained in plant and human cells, little is known about the mechanism of formation of these derivatives in vivo and their potential effects on cell metabolism. Here, we show that NAD(P)HX dehydratase deficiency in yeast leads to an important, temperature-dependent NADHX accumulation in quiescent cells with a concomitant depletion of intracellular NAD+ and serine pools. We demonstrate that NADHX potently inhibits the first step of the serine synthesis pathway in yeast. Human cells deficient in the NAD(P)HX dehydratase also accumulated NADHX and showed decreased viability. In addition, those cells consumed more glucose and produced more lactate, potentially indicating impaired mitochondrial function. Our results provide first insights into how NADHX accumulation affects cellular functions and pave the way for a better understanding of the mechanism(s) underlying the rapid and severe neurodegeneration leading to early death in NADHX repair deficient children. Machine learning has been successfully applied to the prediction of chemical properties of small organic molecules such as energies or polarizabilities. Compared to these properties, the electronic excitation energies pose a much more challenging learning problem. Here, we examine the applicability of two existing machine learning methodologies to the prediction of excitation energies from time-dependent density functional theory. To this end, we systematically study the performance of various 2- and 3-body descriptors as well as the deep neural network SchNet to predict extensive as well as intensive properties such as the transition energies from the ground state to the rst and second excited state. As perhaps expected current state-of-the-art machine learning techniques are more suited to predict extensive as opposed to intensive quantities. We speculate on the need to develop global descriptors that can describe both extensive and intensive properties on equal footing. [less ▲]

The large scale parallel and distributed platforms produce a continuously increasing amount of data which have to be stored, exchanged and used by various jobs allocated on different nodes of the platform. The management of this huge communication demand is crucial for the performance of the system. Meanwhile, we have to deal with more interferences as the trend is to use a single all-purpose interconnection network. In this paper, we consider two different types of communications: the flows induced by data exchanges during computations and the flows related to Input/Output operations. We propose a general model for interference-aware scheduling, where explicit communications are replaced by external topological constraints. Specifically, we limit the interferences of both communication types by adding geometric constraints on the allocation of jobs into machines. The proposed constraints reduce implicitly the data movements by restricting the set of possible allocations for each job. We present this methodology on the case study of simple network topologies, namely the line and the ring. We propose theoretical lower and upper bounds under different assumptions with respect to the platform and jobs characteristics. The obtained results illustrate well the difficulty of the problem even on simple topologies. [less ▲]

OBJECTIVE: In patients with mitochondrial DNA (mtDNA) maintenance disorders and with aging, mtDNA deletions sporadically form and clonally expand within individual muscle fibers, causing respiratory chain deficiency. This study aimed to identify the sub-cellular origin and potential mechanisms underlying this process. METHODS: Serial skeletal muscle cryosections from patients with multiple mtDNA deletions were subjected to subcellular immunofluorescent, histochemical, and genetic analysis. RESULTS: We report respiratory chain-deficient perinuclear foci containing mtDNA deletions, which show local elevations of both mitochondrial mass and mtDNA copy number. These subcellular foci of respiratory chain deficiency are associated with a local increase in mitochondrial biogenesis and unfolded protein response signaling pathways. We also find that the commonly reported segmental pattern of mitochondrial deficiency is consistent with the three-dimensional organization of the human skeletal muscle mitochondrial network. INTERPRETATION: We propose that mtDNA deletions first exceed the biochemical threshold causing biochemical deficiency in focal regions adjacent to the myonuclei, and induce mitochondrial biogenesis before spreading across the muscle fiber. These subcellular resolution data provide new insights into the possible origin of mitochondrial respiratory chain deficiency in mitochondrial myopathy. [less ▲]

The number of smart devices connected to the Internet is growing at an enormous pace and will reach 30 billion within the next five years. A large fraction of these devices have limited processing capabilities and energy supply, which makes the execution of computation-intensive cryptographic algorithms very costly. This problem is exacerbated by the fact that basic optimization techniques like loop unrolling can not (always) be applied since cryptographic software for the IoT often needs to meet strict constraints on code size to not exceed the program storage capacity of the target device. In this paper we introduce SECCCM3, a "lightweight" software library for scalable elliptic curve cryptography on ARM Cortex-M3 microcontrollers. The current version of SECCCM3 is able to carry out variable-base scalar multiplication on Montgomery-form curves over pseudo-Mersenne prime fields, such as Curve25519, and can be used to implement static ECDH key exchange. SECCCM3 is scalable in the sense that it supports curves of different order (as long as certain conditions are met), thereby enabling trade-offs between security and execution time (resp. energy dissipation). We made an effort to protect the field arithmetic against Timing Attacks (TAs) and Simple Power Analysis (SPA), taking into account the so-called early-termination effect of the Cortex-M3 integer multiplier, which makes the latency of "long" multiply instructions operand-dependent. Our experiments show that the integration of countermeasures against information leakage caused by this effect increases the execution time by 34%, while the code size grows by 13%. A TA and SPA-resistant scalar multiplication on Curve25519 has an execution time of 4.565 million clock cycles and consumes approximately 5.1 mJ of energy when executed on a STM32L152RE Cortex-M3 microcontroller. SECCCM3 has a binary code size of 4.0 kB, which includes domain parameters for curves over 159, 191, 223, and 255-bit prime fields. [less ▲]

This paper proposes a Dynamic Demand Estimation (DODE) framework that explicitly accounts for activity scheduling and duration. By assuming a Utility-Based departure time choice model, the time-dependent OD flow becomes a function, whose parameters are those of the utility function(s) within the departure time choice model. In this way, the DODE is solved using a parametric approach, which, on one hand, has less variables to calibrate with respect to the classical bi-level formulation while, on the other hand, it accounts for different trip purposes. Properties of the model are analytically and numerically discussed, showing that the model is more suited for estimating the systematic component of the demand with respect to the standard GLS formulation. Differently from similar approaches in literature, which rely on agent-based microsimulators and require expensive survey data, the proposed framework is applicable with all those DTA models, which are based on OD matrix, and do not necessarily need any data at user level. This has been proven by applying the proposed approach with a standard macroscopic realistic Dynamic Traffic Assignment (DTA) [less ▲]

Service Level Agreements (SLAs) are defining the quality of the services delivered from the Cloud Services Providers (CSPs) to the cloud customers. The services are delivered on a pay-per-use model. The quality of the provided services is not guaranteed by the SLA because it is just a contract. The developments around mobile cloud computing and the advent of edge computing technologies are contributing to the diffusion of the cloud services and the multiplication of offers. Although the cloud services market is growing for the coming years, unfortunately, there is no standard mechanism which exists to verify and assure that delivered services satisfy the signed SLA agreement in an automatic way. The accurate monitoring and modelling of the provided Quality of Service (QoS) is also missing. In this context, we aim at offering an automatic framework named PRESENCE, to evaluate the QoS and SLA compliance of Web Services (WSs) offered across several CSPs. Yet unlike other approaches, PRESENCE aims at quantifying in a fair and by stealth way the performance and scalability of the delivered WS. This article focuses on the first experimental results obtained on the accurate modelisation of each individual performance metrics. Indeed, 19 generated models are provided, out of which 78.9% accurately represent the WS performance metrics for two representative SaaS web services used for the validation of the PRESENCE approach. This opens novel perspectives for assessing the SLA compliance of Cloud providers using the PRESENCE framework. [less ▲]

Eulerian-Lagrangian couplings are nowadays widely used to address engineering and technical problems. In particular, CFD-DEM couplings have been successfully applied to study several configurations ranging from mechanical, to chemical and environmental engineering. However, such simulations are normally very computationally intensive, and the execution time represents a major issue for the applicability of this numerical approach to complex scenarios. With this work, we introduce a novel coupling approach aiming at improving the performance of the parallel CFD-DEM simulations. This strategy relies on two points. First, we propose a new partition-collocation strategy for the parallel execution of CFD–DEM couplings, which can considerably reduce the amount of inter-process communication between the CFD and DEM parts. However, this strategy imposes some alignment constraints on the CFD mesh. Secondly, we adopt a dual-grid multiscale scheme for the CFD-DEM coupling, that is known to offer better numerical properties, and that allows us to obtain more flexibility on the domain partitioning overcoming the alignment constraints. We assess the correctness and performance of our approach on elementary benchmarks and at a large scale with a realistic test-case. The results show a significant performance improvement compared to other state-of-art CFD-DEM couplings presented in the literature. [less ▲]
