Die Vibrissen von Seehunden zeichnen sich durch eine komplizierte dreidimensionale Struk- turierung aus. Sie versetzen den Seehund in die Lage, hydrodynamische Nachlaufspuren von Beutefischen zu detektieren. Mit Hilfe von Stereo- μ Piv Messungen an Seehundvibrissen konnte die stationäre Strömungstopologie des Vibrissennachlaufes ermittelt werden. Zum besseren Verständnis der Funktion der Vibrissenstrukturierung ist es notwendig, die instatio- näre Strömungstopologie zu kennen. Durch räumlich und zeitlich hochauflösende numeri- sche Verfahren konnte die instationäre Strömungstopologie im Nachlauf einer Seehund- vibrisse ermittelt werden. [less ▲]

Due to the rising cost pressures in the construction industry, it is particularly important to choose efficient design methods to achieve economic structures. The use of the plastic hinge theory may lead to good results within the steel and composite construction industry because cross section- and system reserves can almost fully used. This means for the normal composite beam a redistribution of the bending moment from the support section to the field section. This redistribution causes an increase of the rotation at the composite connection. The presented tests show the influence of the reinforcement (type of reinforcement and reinforcement ratio) to the rotation capacity of the connection. The increased rotation requirements of composite connections in the area of plastic hinges should already be take in to account at planning state. [less ▲]

Untraceable communication is about hiding the identity of the sender or the recipient of a message. Currently most systems used in practice (e.g., TOR) rely on the principle that a message is routed via several relays to obfuscate its path through the network. However, as this increases the end-to-end latency it is not ideal for applications like Voice-over-IP (VoIP) communication, where participants will notice annoying delays if the data does not arrive fast enough. We propose an approach based on the paradigm of Dining Cryptographer networks (DC-nets) that can be used to realize untraceable communication within small groups. The main features of our approach are low latency and resilience to packet-loss and fault packets sent by malicious players. We consider the special case of VoIP communication and propose techniques for a P2P implementation. We expose existing problems and sketch possible future large-scale systems composed of multiple groups. [less ▲]

The Janus kinases, Jaks, constitutively associate with the cytoplasmic region of cytokine receptors and play an important role in a multitude of biological processes. Jak2 dysfunction has been implicated in myeloproliferative diseases and leukemia. Although Jaks were studied extensively for many years, the molecular mechanism of Jak activation upon cytokine stimulation of cells is still incompletely understood. In this study, we investigated the importance of an unusual insertion located within the kinase domain in Jak2. We found that the deletion of this insertion, which we named the Jak-specific insertion (JSI), totally abrogates Jak2 autophosphorylation. We further point mutated four residues within the JSI that are conserved in all Jak family members. Three of these mutants showed abrogated or reduced autophosphorylation, whereas the fourth displayed increased autophosphorylation. We found that the phosphorylation state of these mutants is not influenced by other domains of the kinase. Our data further suggest that the JSI is not required for the negative regulation of kinase activity by the suppressor of cytokine signaling proteins, SOCS. Most importantly, we show that mutations in this region differentially affect IFN-gamma and erythropoietin signal transduction. Taken together, the dramatic effects on the phosphorylation status of Jak2 as well as the differential effects on the signaling via different cytokines highlight the importance of this unusual region for the catalytic activity of Jaks. BACKGROUND: The purpose of the study was to identify and molecularly characterize a neurological syndrome in a consanguineous Pakistani family. METHODS: Five patients, their 2 siblings, and their parents were clinically examined. DNA from all 7 siblings was genotyped with Affymetrix SNP arrays and sequencing of selected candidate genes. RESULTS: An unusual neurological syndrome of crawling gait, predominant leg dystonia, pyramidal signs, microcephaly, and suspected deafness segregated in the family. Three patients ambulated on hands and knees, either by hopping and crossing their legs, or by dragging the legs behind them. Two patients have acquired the ability to walk bipedally with a dystonic gait. Unexpectedly, no chromosomal region was homozygous in patients only. Under different disease models, we localized 7 chromosomal regions in the genome common to all patients. No pathogenic mutations were identified in selected candidate genes or the mitochondrial genome. CONCLUSION: We describe an unusual movement disorder syndrome reminiscent of but distinct from Uner Tan syndrome. [less ▲]
