This study examined psychophysiological stress responses to idiosyncratically relevant stress in bulimia nervosa (BN) and binge eating disorder (BED), in relation to autonomic cardiac control and nutritional status. A total of 81 women with BN, BED and healthy controls (HC) took part in an in sensu exposure to idiosyncratic stress. Psychological and peripheral physiological parameters were measured, and tonic heart rate variability, nutritional status, and types of stress were determined. In response to stress exposure, both eating disordered groups showed a stronger reactivity of sadness, and the BED group showed a stronger reactivity of insecurity than the HC group. Desire to binge was increased in the context of interpersonal stress. Stress exposure led to increased cardiovascular activity and reduced electrodermal activity that did not differ by group. The BN-specific symptomatology moderated the association between autonomic cardiac control and psychophysiological stress responses. The results suggest common and specific psychophysiological processes in symptom maintenance through life stress in BN and BED. [less ▲]

Interozeption – die Wahrnehmung von Körpersignalen – spielt bei vielen psychologischen Prozessen eine wichti-ge Rolle, z.B. beim subjektiven Erleben von Emotionen oder bei der Symptomgenese körperbezogener psychi-scher Störungen. Neben Herzschlagdetektionstests, die als Indikator für die interozeptive Genauigkeit heran-gezogen werden, existieren außerdem einige psychophy-siologische Methoden für die Erfassung interozeptiver Prozesse, wie Herzschlag-evozierte Potenziale, respira-torische Widerstände oder die Kopplung von Stimulus-präsentation an den kardialen Zyklus. Ziel dieses Sym-posiums ist es, neue Erkenntnisse zu psychophysiolo-gischen Mechanismen von Interozeption zu präsentieren, die auf unterschiedliche viszerale Organe zurückgehen, sowie deren Integration in normales und pathologisches Erleben und Verhalten zu diskutieren. Zunächst wird Thomas M. Schilling (Universität Trier) den Einfluss von baro-afferenter neuronaler Signalüber-mittlung auf die Integration multisensorischer Stimuli diskutieren. Daraufhin präesentiert Sibylle Petersen (KU Leuven) Ergebnisse zu Effekten von Kategorisierung von Atemwiderständen auf Wahrnehmung und Atem-verhalten, sowie Zusammenhang dieser Effekte mit Furcht vor interozeptiven Reizen. Zoé van Dyck (Uni-versität Luxemburg) wird anschließend einen Vortrag zum Einfluss von kurzzeitiger Nahrungsdeprivation auf die Herzschlagdetektionsgenauigkeit halten. Von Laura Müller (Universitätsklinikum Heidelberg) werden Daten zu Herzschlag-evozierten Potenzialen als Indikatoren für die kortikale Verarbeitung afferenter Signale aus dem kardiovaskulären System bei Boderline-Persönlichkeits-störung im Vergleich zu Gesunden präsentiert. Zum Abschluss diskutiert André Schulz (Universität Luxem-burg) Konvergenzen und Divergenzen von interozepti-ven Selbstberichten, Herzschlagdetektionsgenauigkeit und Herzschlag-evozierter Potenziale bei Depersonalisa-tions-/Derealisationsstörung im Vergleich zu Gesunden. gaming disorder (IGD) has been related to a wide range of detrimental psychological and health consequences. The purpose of the present pilot study was to test the direct and indirect relationships between IGD and emotional trauma, body image dissatisfaction, social anxiety, loneliness, depression, and self-esteem. A total of 242 online gamers completed a survey comprising a comprehensive battery of psychometric self-report scales concerning aforementioned variables. Results indicated that IGD was significantly correlated with all the variables except for body image dissatisfaction. Path analysis indicated an indirect relationship between childhood emotional trauma and IGD through depressive symptoms, while adjusting for gender, age, and number of hours gaming. The findings of the present study indicate that online gamers with a history of emotional abuse and/or neglect have higher levels of depressive symptoms, and that depressive symptoms are important risk factors of IGD. BACKGROUND: In Luxembourg, the extensive phenomenon of school dropout is a prime policy concern in the light of individual, social and economic consequences. Although the authorities report an overall decrease of the national dropout rate, the proportion of early school leavers who remain without any specific occupation is still alarming. Therefore, this study intends a shift of focus from system-inherent to individual factors, including mental health and family correlates, to provide a more comprehensive analysis of the dropout phenomenon. METHODS/DESIGN: The objectives of this study are to investigate the type and prevalence of psychiatric disorders among school dropouts and to compare the findings with those by a matched control group of regularly enrolled students. Furthermore, family variables and socioeconomic status will be analysed, as they are factors likely to interfere with both educational attainment and mental health. A trained psychologist will use structured interviews and self-report forms to investigate for mental health issues, information on schooling, socioeconomic situation and family life. Controls will be matched for gender, age, school type and educational grade. DISCUSSION: As school dropouts face a serious risk of long term professional and social marginalization, there is an evident need for action. Identifying psychosocial risk and protective factors of school dropout will deliver solid insight on how to conceive public health strategies for young people who may need a more customized support to carry out their academic potential. Previous results from our laboratory suggest a combined effect of anger-suppression and family history of cardiovascular disorders in determining cardiovascular responses to mental stress. The present study was designed to determine the effect sizes in cardiovascular reactivity associated with biological risk, psychological risk and the combination of these risk factors using meta-analytical techniques. Results from three independent studies with almost identical experimental procedures provided the basis for the calculation of d, the difference between the means of two groups, divided by the pooled within-group standard deviation. Effect sizes were calculated for the comparison of high versus low biological hypertension risk, high versus low anger suppression, and high versus low combined risk. The results show the largest effect sizes for the comparison of high versus low combined risk. The effect sizes associated with the combination of risk factors were larger than the sum of the effect sizes associated with either factor alone. We conclude that the combination of biological and psychological risk factors in determining cardiovascular reactivity to mental stress is more than the sum of its parts. These findings are discussed in terms of a better understanding of the over-additive effects of multiple cardiovascular risk factors on cardiovascular morbidity and mortality. [less ▲]

Protein topologies can be derived at different description levels considering alpha- and beta-structures. The on-line search tool is based on an object-relational database and provides a query browser for data interrogation by string patterns, keyword queries and sequence similarity. With growing amount of experimental data, the number of known protein structures also increases continuously. Classification of protein structures helps to understand relationships between protein structure and function. The main classification methods based on secondary structures are SCOP, CATH and TOPS, which all classify under different aspects, and therefore can lead to different results. We developed a mathematically unique representation of protein structure topologies at a higher abstraction level providing new aspects of classification and enabling for a fast search through the data. Protein Topology Graph Library (PTGL; http://ptgl.zib.de) aims at providing a database on protein secondary structure topologies, including search facilities, the visualization as intuitive topology diagrams as well as in the 3D structure, and additional information. Secondary structure-based protein topologies are represented uniquely as undirected labeled graphs in four different ways allowing for exploration under different aspects. The linear notations, and the 2D and 3D diagrams of each notation facilitate a deeper understanding of protein topologies. Several search functions for topologies and sub-topologies, BLAST search possibility, and links to SCOP, CATH and PDBsum support individual and large-scale investigation of protein structures. Currently, PTGL comprises topologies of 54,859 protein structures. Main structural patterns for common structural motifs like TIM-barrel or Jelly Roll are pre-implemented, and can easily be searched. BACKGROUND: The protein tyrosine phosphatase nonreceptor type 2 (PTPN22) has been established as a type 1 diabetes susceptibility gene. A recent study found the C1858T variant of this gene to be associated with lower residual fasting C-peptide levels and poorer glycemic control in patients with type 1 diabetes. We investigated the association of the C1858T variant with residual beta-cell function (as assessed by stimulated C-peptide, proinsulin and insulin dose-adjusted HbA1c), glycemic control, daily insulin requirements, diabetic ketoacidosis (DKA) and diabetes-related autoantibodies (IA-2A, GADA, ICA, ZnT8Ab) in children during the first year after diagnosis of type 1 diabetes. METHODS: The C1858T variant was genotyped in an international cohort of children (n = 257 patients) with newly diagnosed type 1 diabetes during 12 months after onset. We investigated the association of this variant with liquid-meal stimulated beta-cell function (proinsulin and C-peptide) and antibody status 1, 6 and 12 months after onset. In addition HbA1c and daily insulin requirements were determined 1, 3, 6, 9 and 12 months after diagnosis. DKA was defined at disease onset. RESULTS: A repeated measurement model of all time points showed the stimulated proinsulin level is significantly higher (22%, p = 0.03) for the T allele carriers the first year after onset. We also found a significant positive association between proinsulin and IA levels (est.: 1.12, p = 0.002), which did not influence the association between PTPN22 and proinsulin (est.: 1.28, p = 0.03). CONCLUSIONS: The T allele of the C1858T variant is positively associated with proinsulin levels during the first 12 months in newly diagnosed type 1 diabetes children. [less ▲]

Computer-aided diagnosis and stratification of COVID-19 based on chest X-ray suffers from weak bias assessment and limited quality-control. Undetected bias induced by inappropriate use of datasets, and improper consideration of confounders prevents the translation of prediction models into clinical practice. By adopting established tools for model evaluation to the task of evaluating datasets, this study provides a systematic appraisal of publicly available COVID-19 chest X-ray datasets, determining their potential use and evaluating potential sources of bias. Only 9 out of more than a hundred identified datasets met at least the criteria for proper assessment of the risk of bias and could be analysed in detail. Remarkably most of the datasets utilised in 201 papers published in peer-reviewed journals, are not among these 9 datasets, thus leading to models with a high risk of bias. This raises concerns about the suitability of such models for clinical use. This systematic review highlights the limited description of datasets employed for modelling and aids researchers to select the most suitable datasets for their task. [less ▲]
