Gait and balance disturbances typically emerge in advanced Parkinson's disease with generally limited response to dopaminergic medication and subthalamic nucleus deep brain stimulation. Therefore, advanced programming with interleaved pulses was put forward to introduce concomittant nigral stimulation on caudal contacts of a subthalamic lead. Here, we hypothesized that the combined stimulation of subthalamic nucleus and substantia nigra pars reticulata improves axial symptoms compared with standard subthalamic nucleus stimulation. Twelve patients were enrolled in this 2 x 2 cross-over double-blind randomized controlled clinical trial and both the safety and efficacy of combined subthalamic nucleus and substantia nigra pars reticulata stimulation were evaluated compared with standard subthalamic nucleus stimulation. The primary outcome measure was the change of a broad-scaled cumulative axial Unified Parkinson's Disease Rating Scale score (Scale II items 13-15, Scale III items 27-31) at '3-week follow-up'. Secondary outcome measures specifically addressed freezing of gait, balance, quality of life, non-motor symptoms and neuropsychiatric symptoms. For the primary outcome measure no statistically significant improvement was observed for combined subthalamic nucleus and substantia nigra pars reticulata stimulation at the '3-week follow-up'. The secondary endpoints, however, revealed that the combined stimulation of subthalamic nucleus and substantia nigra pars reticulata might specifically improve freezing of gait, whereas balance impairment remained unchanged. The combined stimulation of subthalamic nucleus and substantia nigra pars reticulata was safe, and of note, no clinically relevant neuropsychiatric adverse effect was observed. Patients treated with subthalamic nucleus and substantia nigra pars reticulata stimulation revealed no 'global' effect on axial motor domains. However, this study opens the perspective that concomittant stimulation of the substantia nigra pars reticulata possibly improves otherwise resistant freezing of gait and, therefore, highly warrants a subsequent phase III randomized controlled trial. The moose (Alces alces) is a ruminant that harvests energy from fiber-rich lignocellulose material through carbohydrate-active enzymes (CAZymes) produced by its rumen microbes. We applied shotgun metagenomics to rumen contents from six moose to obtain insights into this microbiome. Following binning, 99 metagenome-assembled genomes (MAGs) belonging to 11 prokaryotic phyla were reconstructed and characterized based on phylogeny and CAZyme profile. The taxonomy of these MAGs reflected the overall composition of the metagenome, with dominance of the phyla Bacteroidetes and Firmicutes. Unlike in other ruminants, Spirochaetes constituted a significant proportion of the community and our analyses indicate that the corresponding strains are primarily pectin digesters. Pectin-degrading genes were also common in MAGs of Ruminococcus, Fibrobacteres and Bacteroidetes and were overall overrepresented in the moose microbiome compared with other ruminants. Phylogenomic analyses revealed several clades within the Bacteriodetes without previously characterized genomes. Several of these MAGs encoded a large numbers of dockerins, a module usually associated with cellulosomes. The Bacteroidetes dockerins were often linked to CAZymes and sometimes encoded inside polysaccharide utilization loci, which has never been reported before. The almost 100 CAZyme-annotated genomes reconstructed in this study provide an in-depth view of an efficient lignocellulose-degrading microbiome and prospects for developing enzyme technology for biorefineries. The mammalian gene Nit1 (nitrilase-like protein 1) encodes a protein that is highly conserved in eukaryotes and is thought to act as a tumor suppressor. Despite being ∼35% sequence identical to ω-amidase (Nit2), the Nit1 protein does not hydrolyze efficiently α-ketoglutaramate (a known physiological substrate of Nit2), and its actual enzymatic function has so far remained a puzzle. In the present study, we demonstrate that both the mammalian Nit1 and its yeast ortholog are amidases highly active toward deaminated glutathione (dGSH; i.e., a form of glutathione in which the free amino group has been replaced by a carbonyl group). We further show that Nit1-KO mutants of both human and yeast cells accumulate dGSH and the same compound is excreted in large amounts in the urine of Nit1-KO mice. Finally, we show that several mammalian aminotransferases (transaminases), both cytosolic and mitochondrial, can form dGSH via a common (if slow) side-reaction and provide indirect evidence that transaminases are mainly responsible for dGSH formation in cultured mammalian cells. Altogether, these findings delineate a typical instance of metabolite repair, whereby the promiscuous activity of some abundant enzymes of primary metabolism leads to the formation of a useless and potentially harmful compound, which needs a suitable “repair enzyme” to be destroyed or reconverted into a useful metabolite. The need for a dGSH repair reaction does not appear to be limited to eukaryotes: We demonstrate that Nit1 homologs acting as excellent dGSH amidases also occur in Escherichia coli and other glutathione-producing bacteria. We present a Nitche’s method to couple non-conforming two and three-dimensional NURBS (Non Uniform Rational B-splines) patches in the context of isogeometric analysis (IGA). We present results for linear elastostatics in two and and three-dimensions. The method can deal with surface-surface or volume-volume coupling, and we show how it can be used to handle heterogeneities such as inclusions. We also present preliminary results on modal analysis. This simple coupling method has the potential to increase the applicability of NURBS-based isogeometric analysis for practical applications. [less ▲]

A Nitche’s method is presented to couple different mechanical models. They include coupling of a solid and a beam and of a solid and a plate. Both conforming and non-conforming formulations are presented. In a non-conforming formulation, the structure domain is overlapped by a refined solid model which is needed to either get more accuracy or to capture highly nonlinear events. Applications can be found in multi-dimensional analyses in which parts of a structure are modeled with solid elements and others are modeled using a coarser model with beam and/or plate elements. Discretisations are performed using both standard Lagrange elements and high order NURBS (Non Uniform Rational Bsplines) based isogeometric elements. Un peu plus d’un quart (27%) de la population âgée d’au moins 15 ans ne poursuivant plus d’études possèdent un diplôme de l’enseignement supérieur. Ce pourcentage est un peu plus élevé chez les hommes que chez les femmes (29.7% contre 24.5%). En revanche, un cinquième de la population (19.9%) n’a pas dépassé le niveau d’enseignement primaire. La part des personnes ayant atteint le niveau du secondaire inférieur est de 14.6% et 35.5% de la population ont atteint un niveau d’éducation du secondaire supérieur. Depuis 2001, une augmentation du niveau d’éducation est observée. La part des diplômés de l’enseignement supérieur passe de 19.6% en 2001 à 27.0% en 2011. Les générations les plus âgées ont un niveau d’éducation moins élevé que les générations plus jeunes : 53.5% des 85 ans et plus ont, au maximum, atteint le niveau d’enseignement primaire, contre seulement 9.4% des 20 à 39 ans. Les personnes de nationalité étrangère sont surreprésentées pour deux niveaux d’éducation : le primaire et le supérieur. 24.4% des étrangers n’ont atteint que le niveau d’éducation primaire, contre 16.8% des Luxembourgeois. En revanche 31.8% des étrangers ont atteint un niveau d’éducation supérieur, contre seulement 23.7% des Luxembourgeois. Le type de diplôme obtenu à une grande influence sur la profession exercée. 88.6% des personnes qui travaillent dans les professions intellectuelles et scientifiques ont un diplôme de l’enseignement supérieur, alors qu’ils ne représentent que 1.8% des personnes exerçant une profession élémentaire. À noter également que 57.4% des personnes nées au Grand-Duché et ayant obtenu un diplôme supérieur ont acquis leur diplôme à l’étranger. Si l’on ne prend en compte que les personnes ayant obtenu un master ou un doctorat, ce pourcentage atteint les 90.7%. [less ▲]

This paper introduces two new nominal NK Landscape model instances designed to mimic the properties of one challenging optimisation problem from biology: the Inverse Folding Problem (IFP), here focusing on a simpler secondary structure version. Through landscape analysis tests, numerous problem properties are identified and used to parameterise and validate model instances in terms of epistatic links, adaptive- and random walk characteristics. Then the performance of different Genetic Algorithms (GAs) is compared on both the new NK Models and the original IFP, in terms of population diversity, solution quality and convergence characteristics. It is demonstrated that very similar properties are captured in all presented tests with a significantly faster evaluation time compared to the real IFP. The future purpose of such a model is to provide a generic benchmark for algorithms targeting protein sequence optimisation, specifically in protein design. It may also provide the foundation for more in-depth studies of the size, shape and characteristics of the solution space of good solutions to the IFP. [less ▲]
