Diese Arbeit untersucht zwei Herzschlagwahrnehmungsaufgaben mit unterschiedlichen Verarbeitungsmodi - Heartbeat Tracking Task (HTT; Schandry, 1981) und adaptierter Heartbeat Tracking Task (aHTT; adaptiert nach Melloni et al., 2013) - welche Interozeptive Genauigkeit (IG) anhand des Vergleichs von objektiver und selbst-berichteter Herzrate erfassen (Garfinkel et al., 2015). Die Paradigmen unterscheiden sich hinsichtlich des mentalen Zählens (HTT) und motorischen Tappings (aHTT) von kardiovaskulären Empfindungen. Tapping wird in Biofeedback-Studien zur Verbesserung der IG angewendet (Ring et al., 2015), bislang jedoch kaum mit mentalem Zählen verglichen. 74 gesunde Versuchspersonen (MAlter = 22.66, SDAlter = 2.74, 49 Frauen) bearbeiteten den HTT und den aHTT. Die IGScores des HTT waren im Durchschnitt signifikant höher als im aHTT (t(73) = 2.73, p = .01, d = .28). Bei einem Cut-Off von IG = 0.70 konnten 37 gute Herzschlagwahrnehmer/innen im HTT und 27 gute Herzschlagwahrnehmer/innen im aHTT klassifiziert werden. Auch im Selbstbericht zur eigenen Herzschlagwahrnehmungsfähigkeit, zeigte sich bei den Probanden ein höheres Konfidenzrating im HTT als im aHTT (MHTT = 5.00, SD = 2.04; MaHTT = 3.88, SD = 1.77; Z(74) = -5.24, p < .01, d = .59). Zusammenfassend weisen die Ergebnisse auf eine bessere Herzschlagwahrnehmung im HTT hin, sowohl bezüglich der IG-Scores als auch im Selbstbericht. Eine Erklärung für die niedrigeren Interozeptionswerte im aHTT könnte sein, dass die sensorische und motorische Interferenz der Tapping-Aufgaben mit der Detektion von afferenten Herzsensationen konkurrieren, weshalb der salientere Tapping-Stimulus den Aufmerksamkeitsfokus von internalen Prozessen ablenkt (Pennebaker & Hoover, 1984). Zukünftige Studien sollten prüfen, welche spezifischen Einsatzmöglichkeiten sich für HTT und aHTT anbieten, hinsichtlich einer akkuraten und ökonomischen Erfassung von IG. [less ▲]

Interozeption umfasst die Repräsentation, Verarbeitung und Wahrnehmung körpereigener Prozesse. Veränderungen der Interozeption sind potenziell transdiagnostische Störungsmechanismen bei Psychopathologie. Bisherige Studien deuten auf ein komplexes Zusammenspiel zwischen interozeptiven Prozessen und modulierenden Faktoren, wie humoralen Signalwegen und sozialen Faktoren, hin. Dieses Symposium präsentiert und diskutiert aktuelle Befunde zu pharmakologischen und neurophysiologischen Grundlagen von Interozeption, sowie Interventionen bei Populationen mit psychischen Störungen. Die Auswirkungen von akutem Stress auf die interozeptive Verarbeitung werden von Greta Hansen (Luxemburg) beschrieben. André Schulz (Luxemburg) wird Daten zu gastrisch-interozeptiven Prozessen bei Patienten mit entzündlichen Darmerkrankungen und Reizdarmsyndrom vorstellen. Aline Tiemann (Fribourg) wird Zusammenhänge von gastrischer und kardialer Interozeption mit pathologischem Essverhalten (einschließlich bei Individuen mit Bulimieund Binge-Eating-Symptomatik) vorstellen, sowie Pilotdaten über ein neu entwickeltes virtuelles gastrisches Biofeedback-Paradigma als Intervention bei beeinträchtigter gastrischer Interozeption. Mögliche Interventionen für veränderte neurobiologische Mechanismen im Zusammenhang mit Interozeption umfassen pharmakologische und neurophysiologische Interventionen. Sarah Back (München) wird vagale neurophysiologische Veränderungen im Hinblick auf die Herzfrequenzvariabilität und Herzschlag-evozierte Potenziale bei Borderline-Persönlichkeitsstörung, sowie Modulationen durch das Neuropeptid Oxytocin vorstellen. Schließlich wird Marius Schmitz (Heidelberg) ein ätiologisches Modell interozeptiver Prozesse für Psychopathologie vorstellen und zukünftige Richtungen für das weitere Forschungsfeld aufzeigen. Zusammen zeigen diese Beiträge auf, dass veränderte Interozeption auf verschiedenen Verarbeitungsebenen für spezifische Symptome bei psychischen Störungen verantwortlich sein könnte. Außerdem könnte Interozeption zu diagnostischen Prozessen und als Verlaufsindikator bei Interventionen beitragen. [less ▲]

The generalized Cauchy relation (gCR) of epoxy/silica nanocomposites does not show either the chemically induced sol–gel transition or the chemically induced glass transition in the course of polymerization. Astonishingly, by varying the silica nanoparticles’ concentration between 0 and 25 vol%in the composites, the Cauchy parameter A of the gCR remains universal and can be determined from the pure epoxy’s elastic moduli. Air-filled porous silica glasses are considered as models for percolated silica particles. A longitudinal modulus versus density representation evidences the aforementioned transition phenomena during polymerization of the epoxy/silica nanocomposites. The existence of optically and mechanically relevant interphases is discussed. [less ▲]

Ageing, the most salient policy challenge of the 21st century, can convert into an economic venture. In order to grasp this aspiration, this paper examines the interplay between active ageing and the demand side of the silver economy from both national and the EU perspective. The goal is to configure a combination of policies recommended by the active ageing framework that ameliorates silver consumption. Qualitative comparative analysis (QCA) demonstrated that the intersection between higher public health care spending, active social life, achieving employment targets and retirement before or at statutory retirement age constitutes a sufficient combination to exploit the potential of the silver economy. Therefore, the active ageing framework theoretically underpins the silver. However, the argument is not empirically compelling because only a minor proportion of the EU member states have actively engaged in the exploitation of the silver economy potential. [less ▲]

Many studies have examined the effect of life events, education, and income on well-being. Conversely, research concerning well-being as a predictor of life course outcomes is sparse. Diener’s suggestion “to inquire about the effects of well-being on future behavior and success” has, with some exceptions, not yet come to fruition. This article contributes to this body of research. We conceptualize and analyze the interplay between educational achievement, occupational success, and well-being as a complex process. The relationship between these domains is examined drawing on a structure-agency framework derived from Bourdieu and Social Comparison Theory. Social comparison between adolescents and their parents is suggested to be the mechanism explaining the effects of successful and unsuccessful intergenerational transmission of educational achievement and occupational success on well-being. It is further argued that well-being may serve as an individual resource by fostering educational and occupational outcomes. Panel data from the Transition from Education to Employment (TREE) project, a Swiss PISA 2000 follow-up study, was used. The interplay between well-being and successful and unsuccessful intergenerational transfer of educational attainment was analyzed in an autoregressive cross-lagged mixture model framework. Social comparison was found to be related to well-being, while well-being proved to significantly increase the probability of successful intergenerational transfer of educational attainment. The protein L-isoaspartyl-O-methyltransferase functions to initiate the repair of isomerized aspartyl and asparaginyl residues that spontaneously accumulate with age in a variety of organisms. Caenorhabditis elegans nematodes lacking the pcm-1 gene encoding this enzyme display a normal lifespan and phenotype under standard laboratory growth conditions. However, significant defects in development, egg laying, dauer survival, and autophagy have been observed in pcm-1 mutant nematodes when deprived of food and when exposed to oxidative stress. Interestingly, overexpression of this repair enzyme in both Drosophila and C. elegans extends adult lifespan under thermal stress. In this work, we show the involvement of the insulin/insulin-like growth factor-1 signaling (IIS) pathway in PCM-1-dependent lifespan extension in C. elegans. We demonstrate that reducing the levels of the DAF-16 downstream transcriptional effector of the IIS pathway by RNA interference reduces the lifespan extension resulting from PCM-1 overexpression. Using quantitative real-time PCR analysis, we show the up-regulation of DAF-16-dependent stress response genes in the PCM-1 overexpressor animals compared to wild-type and pcm-1 mutant nematodes under mild thermal stress conditions. Additionally, similar to other long-lived C. elegans mutants in the IIS pathway, including daf-2 and age-1 mutants, PCM-1 overexpressor adult animals display increased resistance to severe thermal stress, whereas pcm-1 mutant animals survive less long under these conditions. Although we observe a higher accumulation of damaged proteins in pcm-1 mutant nematodes, the basal level of isoaspartyl residues detected in wild-type animals was not reduced by PCM-1 overexpression. Our results support a signaling role for the protein L-isoaspartyl methyltransferase in lifespan extension that involves the IIS pathway, but that may be independent of its function in overall protein repair. In particular, we focus on how to implement roles in an abstract way in objects representing relationships, and to specify the interconnections between the roles. Abstract roles cannot be instantiated. To participate in a relationship, objects have to extend the abstract roles of the relationship. Enhanced legitimacy is a driving force behind deliberative innovations (Fung 2015, Curato and Böker 2016). This is no less the case for constitutional deliberative innovations. Assessing deliberative constitutionalism’s success in generating legitimacy necessitates a better grasp of the distinct legitimacy standards which constitutional deliberative innovations may meet. Bound up with those standards is deliberative constitutionalism’s attempt to navigate the tension between politics and law, constituent power (CP) and constituted form (CF). How do factors of deliberative legitimacy interact with standard ways of modelling that tension? To answer this question, I proceed in three parts, the first of which maps ten factors of deliberative legitimacy at four levels: personal, interpersonal, institutional, and systemic. The second lays out four ways of modelling the tension between CP and CF from Loughlin and Walker (2007): the containment, mutual articulation, radical potential, and irritant models. The last part sets out to determine whether the four models directly or indirectly support or neglect the realization of the forms of deliberative legitimacy and casts in a different light from Parkinson (2016) the landscape of deliberative democracy and constitutions. I conclude that certain forms of deliberative legitimacy may be more sensitive to and better served by some models of CP and CF than by others. [less ▲]

Differential effects of measles virus (MV) on the innate immune response may influence virus spread and severity of disease. Using a representative panel of 22 MV strains including 14 different genotypes, we found that wild-type (wt) differ considerably in their sensitivity to type I interferon (IFN). The wt virus production was 2-47-fold lower in IFN-alpha treated Vero/hSLAM cells, whereas vaccine virus production was reduced only 2-3-fold. Sequence analysis of the MV-P/C/V gene, revealed no obvious amino acid mutations that correlated with the different phenotypes. Strains also widely differed in their ability to induce type I IFN, tumor necrosis factor (TNF) alpha and other cytokines in human A549/hSLAM cells. Some wt strains that were highly sensitive to type I IFN induced only low levels of these and other cytokines. In vitro wt strains that produced the 5' copy-back defective interfering RNAs (5'cb-diRNA) characterized by Shingai et al. (2007), induced high levels of cytokines that otherwise were only reached by vaccine strains. These 5'cb-diRNAs emerged only in virus cultures during multiple passaging and were not detectable in clinical samples of measles patients. These subgenomic RNAs are an important confounding parameter in passaged wt viruses which must be carefully assessed in all in vitro studies. The present data show that MV wt strains differ in their sensitivity and their ability to temper with the innate immune response, which may result in differences in virulence. [less ▲]

MicroRNAs (miRNAs) are ubiquitously expressed small non-coding RNAs that, in most cases, negatively regulate gene expression at the posttranscriptional level. miRNAs are involved in fine-tuning fundamental cellular processes such as proliferation, cell death and cell cycle control and are believed to confer robustness to biological responses. Here, we investigated simultaneously the transcriptional changes of miRNA and mRNA expression levels over time after activation of the Janus kinase/Signal transducer and activator of transcription (Jak/STAT) pathway by interferon-c stimulation of melanoma cells. To examine global miRNA and mRNA expression patterns, time-series microarray data were analysed. We observed delayed responses of miRNAs (after 24-48 h) with respect to mRNAs (12-24 h) and identified biological functions involved at each step of the cellular response. Inference of the upstream regulators allowed for identification of transcriptional regulators involved in cellular reactions to interferon-c stimulation. Linking expression profiles of transcriptional regulators and miRNAs with their annotated functions, we demonstrate the dynamic interplay of miRNAs and upstream regulators with biological functions. Finally, our data revealed network motifs in the form of feed-forward loops involving transcriptional regulators, mRNAs and miRNAs. Additional information obtained from integrating time-series mRNA and miRNA data may represent an important step towards understanding the regulatory principles of gene expression. © The Author(s) 2013. Big Five personality traits are assumed to be linked with attitudes toward own aging. Since both constructs have central importance for the aging process, it is surprising that to our knowledge no study so far comprehensively addressed their mutual connection over time. We used data from the ILSE study, a longitudinal study capturing personality and attitudes toward own aging at four measurement occasions, spanning 20 years and including two participant cohorts in midlife (n = 501; born 1950–52) and later life (n = 500; born 1930–32). Dual latent change score models showed that personality was longitudinally related to change in attitudes toward own aging: Lower Neuroticism, higher Conscientiousness, and higher Openness predicted more positive attitudes, whereas the direction of the effect for Extraversion varied by time. Furthermore, the role of personality seems to be confined to certain sensitive periods in midlife and early old age. Contrary to our expectations, attitudes toward own aging had only marginal longitudinal impact on the Big Five. Our results shed light on the developmental co-dynamics of personality and subjective perceptions of aging across the second half of the lifespan. [less ▲]
