Jaks are predicted to have a 4.1, ezrin, radixin, moesin (FERM) domain at their N terminus. FERM domains are composed of three structurally unrelated subdomains (F1, F2, and F3) which are in close contact to one another and form the clover-shaped FERM domain. We generated a model structure of the Jak1 FERM domain, based on solved FERM structures and the alignments with other FERM domains. To destabilize different subdomains and to uncover their exact function, we mutated specific hydrophobic residues conserved in FERM domains and involved in hydrophobic core interactions. In this study, we show that the structural integrity of the F2 subdomain of the FERM domain of Jak1 is necessary to bind the IFN-gammaRalpha. By mutagenesis of hydrophobic residues in the hydrophobic core between the three FERM subdomains, we find that the structural context of the FERM domain is necessary for the inhibition of Jak1 phosphorylation. Thus, FERM domain mutations can have repercussions on Jak1 function. Interestingly, a mutation in the kinase domain (Jak1-K907E), known to abolish the catalytic activity, also leads to an impaired binding to the IFN-gammaRalpha when this mutant is expressed at endogenous levels in U4C cells. Our data show that the structural integrity of both the FERM domain and of the kinase domain is essential for both receptor binding and catalytic function/autoinhibition. [more ▼] This exploratory study is devoted in equal measure to the status quo and the future perspectives of the internationalisation of dual study programmes, a special hybrid form of vocational training and higher education developed in Germany. Building on the earlier DAAD studies ‘Sachstand: Duales Studium als Exportmodell’ (Maschke 2012) and ‘Modelle und Szenarien für den Export deutscher Studienangebote ins Ausland’ (Schreiterer and Witte 2001; see also DAAD/HRK 2012), the authors examine both the degree of internationalisation of existing dual study programmes in Germany (with special emphasis on students’ geographical mobility) and the possibilities and limits of systematically transferring this emergent educational model to selected countries. Two recent trends have helped put issues of internationalisation and the transfer of German education concepts higher up on the policy agenda again: first, the current economic situation in Germany, which has remained robust despite the recent financial and economic turmoil, reflected most importantly in comparatively low levels of youth unemployment. Dual vocational education and training models are seen as a key factor contributing to this success. Second, the concept of dual studies reflects an emergent model of skill formation at the nexus of initial vocational training and tertiary education. This innovative hybrid form is seen as having the potential to play a crucial role in the development of competencies for twenty-first-century occupations, not least against the backdrop of the pressing skills gap. [more ▼] In this paper, we apply non-orthogonal multiple access (NOMA) in satellite systems to assist data transmission for services with latency constraints. We investigate a problem to minimize the transmission time by jointly optimizing power allocation and terminal-timeslot assignment for accomplishing a transmission task in NOMA-enabled satellite systems. The problem appears non-linear/non-convex with integer variables and can be equivalently reformulated in the format of mixed-integer convex programming (MICP). Conventional iterative methods may apply but at the expenses of high computational complexity in approaching the optimum or near-optimum. We propose a combined learning and optimization scheme to tackle the problem, where the primal MICP is decomposed into two learning-suited classification tasks and a power allocation problem. In the proposed scheme, the first learning task is to predict the integer variables while the second task is to guarantee the feasibility of the solutions. Numerical results show that the proposed algorithm outperforms benchmarks in terms of average computational time, transmission time performance, and feasibility guarantee.
