The rheology of dense granular flows is investigated through discrete element method (DEM) simulation of a vane shear cell. From the simulation, profiles of shear stress, shear rate, and velocity are obtained, which demonstrates that the flow features in the vane shear cell are equivalent to those in the classic annular Couette cell. A novel correlation for the shear viscosity is formulated and leads to a new expression for μKT in the kinetic theory analysis. The μKT formulation is able to qualitatively capture the μ-I relation in the shear cell. A correlation length is added in the energy dissipation term to account for the effects of the particle motion correlation. A simplified correlation length model is derived based on DEM results and is compared with the literature. The modified granular kinetic energy equation is able to correctly predict the granular temperature profiles in the shear cell. [less ▲]

Transcription initiation and RNA processing govern gene expression and enable bacterial adaptation by reshaping the RNA landscape. The aim of this study was to simultaneously observe these two fundamental processes in a transcriptome responding to an environmental signal. A controlled σE system in E. coli was coupled to our previously-described tagRNA-seq method to yield process kinetics information. Changes in transcription initiation frequencies (TIF) and RNA processing frequencies (PF) were followed using 5’ RNA tags. Changes in TIF showed a binary increased/decreased pattern that alternated between transcriptionally activated and repressed promoters, providing the bacterial population with transcriptional oscillation. PF variation fell into three categories of cleavage activity; i) constant and independent of RNA levels, ii) increased once RNA has accumulated, and iii) positively correlated to changes in TIF. This work provides a comprehensive and dynamic view of major events leading to transcriptomic reshaping during bacterial adaptation. It unveils an interplay between transcription initiation and the activity of specific RNA cleavage sites. This study utilized a well-known genetic system to analyze fundamental processes, and can serve as blueprint for comprehensive studies that exploit the RNA metabolism to decipher and understand bacterial gene expression control. This paper discusses the added value of applying machine learning (ML) to contextually enrich digital collections. In this study, we employed ML as a method to geographically enrich historical datasets. Specifically, we used a sequence tagging tool (Riedl and Padó 2018) which implements TensorFlow to perform NER on a corpus of historical immigrant newspapers. Afterwards, the entities were extracted and geocoded. The aim was to prepare large quantities of unstructured data for a conceptual historical analysis of geographical references. The intention was to develop a method that would assist researchers working in spatial humanities, a recently emerged interdisciplinary field focused on geographic and conceptual space. Here we describe the ML methodology and the geocoding phase of the project, focussing on the advantages and challenges of this approach, particularly for humanities scholars. We also argue that, by choosing to use largely neglected sources such as immigrant newspapers (a lso known as ethnic newspapers), this study contributes to the debate about diversity representation and archival biases in digital practices. The introductory article deals with this and reconstructs the formation and differentiation of the bordering approach. Furthermore, the concept of border experiences is determined as an investigative perspective that is interested in everyday cultural arenas of bordering processes. It puts the agency of ‘border(lands) residents’ in the center and provides insights into everyday cultural border (re)productions.With this in mind, we will present the book articles in the final section. Moral dilemmas have become increasingly popular in single player video games, although it is still widely unknown why players find them pleasurable, how they process dilemmas, and which variables affect the processing. Therefore, three different focus groups sessions with experienced players (N = 16) were conducted. Player perceptions of meaningful and morally relevant decision situations in video games were grouped for topics and contextualized with theoretical background (e.g., moral disengagement theory). Our findings support the notion that moral decision-making in video games is a dynamic interplay between game and user-dependent variables. Results show that in addition to interactivity, which reflects the inherent property of video games, statements can be broadly grouped into factors that describe player motivation (i.e., why they morally engage or disengage) and influencing factors that shape the moral interaction itself. In summary, the present findings provide insights into players' processing of moral dilemmas in video games. [less ▲]

Early adolescence represents a particularly sensitive period in the life of young learners, which is accompanied by an increase in school alienation. Due to its harmful nature (Hascher and Hadjar in Educ Res 60:171–188, 2018. https ://doi.org/10.1080/00131 881.2018.14430 21), school alienation may lead to unfavorable consequences such as low academic achievement (Johnson in J Educ Technol Soc8:179–189, 2005; Reinke and Herman in Psychol Schools 39:549–559, 2002). This study investigates the longitudinal relationship between school alienation domains, namely alienation from learning, from teachers, and from classmates, and academic achievement among secondary school students of grade 7 to grade 9 in Switzerland and Luxembourg. Data were collected from 403 students in the Swiss canton of Bern and 387 students in Luxembourg who participated in three waves of the “School Alienation in Switzerland and Luxembourg (SASAL)” research project. Cross-lagged modeling was applied to examine the correlations between school alienation domains and academic achievement at each of the three time points, the temporal stability of school alienation domains and academic achievement, and their cross-lagged effects across time, controlling for students’ gender, school track, parental occupational status, and migration background. Results show that the pattern of relationships is defined by the school alienation domain and the cultural context, pointing to the complex interplay between the multidimensional construct of school alienation and academic outcomes of secondary school students. Objective: Criterion A of the alternative model for the classification of personality disorders in the DSM-5 introduced the Level of Personality Functioning Scale (LPFS), a dimensional model for the assessment of impairments in self and interpersonal functioning. The LPFS was developed based on a review of different measures of personality functioning, such as the Reflective Functioning Scale, a measure of mentalizing. This study investigated the empirical overlap between LPFS and mentalization. Methods: The study sample included adult inpatients (n = 55) with a mental disorder and a healthy adult control group (n = 55). All participants were examined regarding the LPFS using the Semi-Structured Interview for Personality Functioning DSM-5 (STiP-5.1); mentalizing was assessed with the Brief Reflective Functioning Interview and coded with the Reflective Functioning Scale. We used structural equation modeling to investigate the relationship between LPFS domains and mentalization. Correlation analysis was used to examine the agreement between interview-rated LPFS and self-report measures of personality dysfunction. Results: All domains of the LPFS were significantly related to mentalizing. Interview-rated LPFS was significantly associated with self-reported personality dysfunction. Conclusion: The findings support the notion that the LPFS and mentalization share a strong conceptual and operational overlap by demonstrating that both constructs are empirically interrelated. The results yield further support for the validity of the LPFS as a dimensional model for the assessment of personality disorder severity. [less ▲]

Abstract Flash flood events are expected to become increasingly common with the global increases in weather extremes. They are a significant natural hazard that affects karst landscapes, which host large resources of drinking water worldwide. The role played by underground flood events in the karst aquifer recharge is complex due to the heterogeneity of the basement which remains poorly understood. We present the analysis of 20 in-cave flash flood events affecting the Rochefort karst system (Belgium) using continuous gravity measurements at one single station, and water level sensors installed in caves. Underground flood events typically produce a peak in the gravity signal, due to an increase in the associated mass change. After the flood, the gravity values drop but remain slightly increased compared to before the flood event. Via forward gravity modeling, we demonstrate that this remaining anomaly can be reasonably explained by the infiltration of local rainfall within the karst system rather than by allogenic recharge of the aquifer. Flash floods are mainly restricted to connected voids. This allows us to utilize them as proxies to investigate the distribution of cavities in the karst system. Forward modeling of the gravitational attraction induced by the mapped caves being flooded yields a gravity signal much smaller than the observed one. We conclude that at least 50 more cavities than those previously mapped are required to match the measured anomalies. This presents opportunities for implementing similar approaches in other diverse porous media, using gravity monitoring of hydrological processes (e.g. infiltration fronts, hydrothermalism or tide effects in coastal aquifers) as proxies to characterize underground properties. Abstract Objective Drug resistance is a major concern in the treatment of individuals with epilepsy. No genetic markers for resistance to individual antiseizure medication (ASM) have yet been identified. We aimed to identify the role of rare genetic variants in drug resistance for three common ASMs: levetiracetam (LEV), lamotrigine (LTG), and valproic acid (VPA). Methods A cohort of 1622 individuals of European descent with epilepsy was deeply phenotyped and underwent whole exome sequencing (WES), comprising 575 taking LEV, 826 LTG, and 782 VPA. We performed gene- and gene set–based collapsing analyses comparing responders and nonresponders to the three drugs to determine the burden of different categories of rare genetic variants. Results We observed a marginally significant enrichment of rare missense, truncating, and splice region variants in individuals who were resistant to VPA compared to VPA responders for genes involved in VPA pharmacokinetics. We also found a borderline significant enrichment of truncating and splice region variants in the synaptic vesicle glycoprotein (SV2) gene family in nonresponders compared to responders to LEV. We did not see any significant enrichment using a gene-based approach. Significance In our pharmacogenetic study, we identified a slightly increased burden of damaging variants in gene groups related to drug kinetics or targeting in individuals presenting with drug resistance to VPA or LEV. Such variants could thus determine a genetic contribution to drug resistance. [less ▲]

The role of twins in research is evolving as we move further into the post-genomic era. With the re-definition of what a gene is, it is becoming clear that biological family members who share a specific genetic variant may well not have a similar risk for future disease. This has somewhat invalidated the prior rationale for twin studies. Case co-twin study designs, however, are slowly emerging as the ideal tool to identify both environmentally induced epigenetic marks and epigenetic disease-associated processes. Here, we propose that twin lives are not as identical as commonly assumed and that the case co-twin study design can be used to investigate the effects of the adult social environment. We present the elements in the (social) environment that are likely to affect the epigenome and measures in which twins may diverge. Using data from the German TwinLife registry, we confirm divergence in both the events that occur and the salience for the individual start as early as age 11. Case co-twin studies allow for the exploitation of these divergences, permitting the investigation of the role of not only the adult social environment, but also the salience of an event or environment for the individual, in determining lifelong health trajectories. In cases like social adversity where it is clearly not possible to perform a randomised-controlled trial, we propose that the case cotwin study design is the most rigorous manner with which to investigate epigenetic mechanisms encoding environmental exposure. The role of the case co-twin design will continue to evolve, as we argue that it will permit causal inference from observational data. [less ▲]

The Ethereum blockchain enables the execution of so-called smart contracts. These are programs that facilitate the automated transfer of funds according to a given business logic without the participants requiring to trust one another. However, recently attackers started using smart contracts to lure users into traps by deploying contracts that pretend to give away funds but in fact contain hidden traps. This new type of scam is commonly referred to as honeypots. In this paper, we propose a system that aims to protect users from falling into these traps. The system consists of a plugin for MetaMask and a back-end service that continuously scans the Ethereum blockchain for honeypots. Whenever a user is about to perform a transaction through MetaMask, our plugin sends a request to the back-end and warns the user if the target contract is a honeypot. [less ▲]
