As applications, we obtain isoperimetric inequalities, using Ledoux's argument, and uniform quantitative gradient estimates, firstly for bounded C^2 functions with boundary conditions and then for the unit spectral projection operators of Dirichlet and Neumann Laplacians. The size and form of cities influence their social and environmental impacts. Whether cities have the same form irrespective of their size is still an open question. We analyse the profile of artificial land and population density, with respect to the distance to their main centre, for the 300 largest European cities. Our analysis combines the GMES/Copernicus Urban Atlas 2006 land use database at 5 m resolution for 300 larger urban zones with more than 100,000 inhabitants and the Geostat population grid at 1 km resolution. We find a remarkable constancy of radial profiles across city sizes. Artificial land profiles scale in the two horizontal dimensions with the square root of city population, while population density profiles scale in three dimensions with its cube root. In short, cities of different size are homothetic in terms of land use and population density, which challenges the idea that larger cities are more parsimonious in the use of land per capita. While earlier literature documented the scaling of average densities (total surface and population) with city size, we document the scaling of the whole radial distance profile with city size, thus liaising intra-urban radial analysis and systems of cities. Our findings also yield homogenous spatial definitions of cities, from which we can re-question urban scaling laws and Zipf’s law for cities. [less ▲]

A multitude of factors contribute to complex diseases and can be measured with ‘omics’ methods. Databases facilitate data interpretation for underlying mechanisms. Here, we describe the Virtual Metabolic Human (VMH, www.vmh.life) database encapsulating current knowledge of human metabolism within five interlinked resources ‘Human metabolism’, ‘Gut microbiome’, ‘Disease’, ‘Nutrition’, and ‘ReconMaps’. The VMH captures 5180 unique metabolites, 17 730 unique reactions, 3695 human genes, 255 Mendelian diseases, 818 microbes, 632 685 microbial genes and 8790 food items. The VMH’s unique features are (i) the hosting of the metabolic reconstructions of human and gut microbes amenable for metabolic modeling; (ii) seven human metabolic maps for data visualization; (iii) a nutrition designer; (iv) a user-friendly webpage and application-programming interface to access its content; (v) user feedback option for community engagement and (vi) the connection of its entities to 57 other web resources. The VMH represents a novel, interdisciplinary database for data interpretation and hypothesis generation to the biomedical community. [less ▲]

While genetic advances have successfully defined part of the complexity in Parkinson’s disease (PD), the clinical characterization of phenotypes remains challenging. Therapeutic trials and cohort studies typically include patients with earlier disease stages and exclude comorbidities, thus ignoring a substantial part of the real-world PD population. To account for these limitations, we implemented the Luxembourg PD study as a comprehensive clinical, molecular and device-based approach including patients with typical PD and atypical parkinsonism, irrespective of their disease stage, age, comorbidities, or linguistic background. To provide a large, longitudinally followed, and deeply phenotyped set of patients and controls for clinical and fundamental research on PD, we implemented an open-source digital platform that can be harmonized with international PD cohort studies. Our interests also reflect Luxembourg-specific areas of PD research, including vision, gait, and cognition. This effort is flanked by comprehensive biosampling efforts assuring high quality and sustained availability of body liquids and tissue biopsies. We provide evidence for the feasibility of such a cohort program with deep phenotyping and high quality biosampling on parkinsonism in an environment with structural specificities and alert the international research community to our willingness to collaborate with other centers. The combination of advanced clinical phenotyping approaches including device-based assessment will create a comprehensive assessment of the disease and its variants, its interaction with comorbidities and its progression. We envision the Luxembourg Parkinson’s study as an important research platform for defining early diagnosis and progression markers that translate into stratified treatment approaches. [less ▲]

Dravet syndrome (DS) is one of the most frequent genetic epilepsies, with an incidence of 1/30,000. Most DS patients are pharmacoresistant, in that they do not respond adequately to currently available anticonvulsant drugs (ASDs). Beside the seizure occurrence in these patients, many DS patients also suffer from cognitive impairment that can be aggravated by some of the seizure medications prescribed. Preclinical models such as mouse models of Dravet syndrome have been developed and described to exhibit cognitive deficits similar to those of DS patients, but are only suitable for the evaluation of small number of compounds, thereby limiting their utility for drug discovery. An animal model with higher screening throughput would therefore be of value for drug discovery efforts focused on seizure reduction and decrease of the comorbidities associated with DS. Recent studies on zebrafish have demonstrated its ability to be a promising in vivo model for DS. Two different zebrafish DS models - one based on a loss-of-function mutation in the zebrafish ortholog of SCN1A, the other based on an antisense knockdown of this gene - exhibit seizure-like behaviour and epileptiform discharges that are exacerbated by hyperthermia. The mutant line was also described to display greater anxiety levels. In this Doctoral thesis project, we are investigating these zebrafish DS models with regard to (1) their seizure occurrence and potential reduction following exposure to several anticonvulsant drug candidates, and (2), their cognitive functions in order to determine possible similarities with cognitive impairment in human DS patients. Our results indicate these zebrafish DS models to exhibit memory impairment and higher anxiety levels. These findings provide an initial insight into the resemblance between human patients and zebrafish in terms of comorbidities. Finally, we also identified several novel anticonvulsant compounds and drug candidates with antiseizure activity in these zebrafish DS models. [less ▲]

Given the current abundance of available digital resources for historians (retro-digitized as well as born-digital materials), there is an urgent question of how to engage with these resources in the history classroom. How do we teach the differences between "online" and "offline" materials and they ways in which historians can approach them? How do we deal with issues of decontextualization and the changes brought about in historical research practices by working with digital objects? The SourceLab project tackles these issues by letting students create digital editions of historical artefacts and thereby engage them in the making of history. The Ranke.2 project provides lessons that teaches students what digital source criticism is. In this workshop we will discuss in what ways teachers use digital resources in the classroom, and what strategies, if any, they use to sensitize their students to the specificities of online materials as opposed to those found in the archive. The outcome could be a working paper which discusses the pros and cons of various approaches. [less ▲]

More and more countries are moving toward adopting inclusive school systems. In this context, research has clearly indicated that in order to ensure the successful implementation of this concept, it is crucial for all people involved in the process of restructuring to have positive attitudes. However, even if many studies on different actors´ attitudes toward inclusive education have already been published, the stance of people in Luxembourg on this topic remains unclear. This dissertation uses the Bioecological Model of Inclusive Education to provide an overview of the environmental factors that play key roles in the implementation of inclusive education, such as the decisions made on a governmental level and the attitudes of the Luxemburgish general population as well as those of pre- and in-service teachers. The materials used in the current studies are based on the Three Component Model of attitudes to investigate attitudes toward the general idea of an inclusive school system and (the inclusion of) students with learning difficulties and challenging behavior. To avoid issues with social desirability, implicit measurement tools were additionally used in the first three studies. An analysis of the recent changes in the Luxemburgish educational system revealed that a range of barriers still persist. In this regard, results have shown that the idea of an inclusive school system is embraced by the Luxemburgish general population. By contrast, attitudes toward (the inclusion of) students with special educational needs tend to be negative. Pre- and in-service teachers’ ratings of student achievement of students with learning difficulties and challenging behavior (in Study 2) were below average and influenced by stereotypical beliefs. However, teachers with experience in inclusive classrooms gave the highest ratings of both groups of students. These findings raised the question of whether, in addition to experience, the definition of inclusive education plays a role in the development of positive attitudes. The results of Study 4 indeed revealed associations between definitions and attitudes, whereby people who emphasized that inclusive education is about valuing the needs of all students held the most positive attitudes. Notably, the individual readiness of teachers to implement inclusive education was also related to the definition. Implications of the findings for the educational system and the society as well as for (attitude) research in the field of inclusive education are outlined, and directions for future research are given. In recent years, tangible user interfaces (TUI) have gained in popularity in educational contexts, among others to implement problem-solving and discovery learning science activities. In the context of an interdisciplinary and cross-institutional collaboration, we conducted a multimodal EMCA-based video user study involving a TUI-mediated bicycle mechanics simulation. This article focusses on the discovering work of a group of three students with regard to a particular tangible object (a red button), designed to support participants engagement with the underlying physics aspects and its consequences with regard to their engagement with the targeted mechanics aspects. 404 not found, closed and opaque corpora, websites disappearance after a few years, obsolete tools, problems of maintenance … this workshop wishes to explore the instability of data, of their digital (digitalized or/and born-digital) heritage, of digital technologies and tools, the numerous transformations of devices, platforms, terms of use and the tensions between this instability and the necessary stability and reproducibility of research. How should researchers deal at the same time with the ephemeral, the flow, the obsolescence and the concern of knowledge production? How is it possible to reconciliate the temporality of research and the short cycles of technologies? This workshop aims at introducing and identifying the challenges of sustainability and durability which are too often underestimated in digital research and rarely presented to students when introducing tools and research methods. This 120 minutes workshop will be organized in three parts as follows: - Identifying the levels, steps, in which sustainability and durability of a digital research are challenged, from the shaping and sharing of corpora to the maintenance of research results and digital productions. This will be based on the participants experiences and on several examples of research projects; - Mapping the issues and existing solutions such as digital research infrastructures, permanent identifiers, licenses, but also obstacles and limits (such as author rights …) that researchers have to face or take into account; - Creating practical guidelines that could be used by scholars and students. This workshop expands a research project conducted by Christine Barats (Céditec, Université Paris- Est Créteil), Andreas Fickers (Head of C2DH, University of Luxembourg), Valérie Schafer (C2DH, University of Luxembourg).
