A large storm surge event occurred on 2007 November 2009 in the southern North Sea where strong winds caused the sea level to rise drastically by up to 3 m within several hours. Based on the Proudman Oceanographic Laboratory storm surge model, the predicted loading displacements at coastal stations can reach a few centimetres in the vertical and several millimetres in the horizontal directions. In this study, we used two-hourly global positioning system (GPS) positions at 26 stations around the southern North Sea to identify the loading displacements caused by this storm surge event. We find that the mean rms of the differences between the estimated and predicted displacements are 4.9, 1.3 and 1.4 mm, which are insignificant compared to the one-sigma GPS positioning errors of 5.1, 2.0 and 2.4 mm for the Up, East and North components, respectively. More interestingly, in both vertical and horizontal directions, the estimated displacements successfully tracked the temporal evolution of the storm surge loading effects. In addition, within the whole of 2007 November, we used the predicted displacements to correct the two-hourly GPS positions, and consequently reduced the rms of the estimated displacements on average from 9.3, 3.0 and 2.9 mm to 7.8, 2.8 and 2.8 mm for Up, East and North components, respectively. Therefore, subdaily loading effects due to storm surges should be paid attention to in the GPS positioning that contributes to crustal-motion studies around shallow seas such as the North Sea, the Baltic Sea and the Gulf of Mexico. Manche Menschen bezeichnen sich als süchtig nach bestimmten Nahrungsmitteln wie Schokolade. Des Weiteren wurden auf Verhaltens- und neurobiologischer Ebene Überschneidungen zwischen Substanzabhängigkeiten und exzessivem Essen wie es bei Bulimia nervosa (BN), Binge Eating Störung (BES) oder Adipositas auftritt, gefunden (Davis & Carter, 2009). Bisher mangelte es im deutschen Sprachraum jedoch an einem Messinstrument, das spezifisch das Konstrukt Esssucht erfasst. Um diese Lücke zu schließen, stellt die vorliegende Arbeit die deutsche Version der Yale Food Addiction Scale (YFAS) von Gearhardt, Corbin & Brownell (2009) vor. Diese Skala ermöglicht es, Menschen mit ausgeprägten Symptomen einer Abhängigkeit von bestimmten Nahrungsmitteln zu identifizieren. Die Diagnosestellung orientiert sich hierbei an den DSM-IV-Kriterien für Substanzabhängigkeit. Die Reliabilität und Validität dieses Fragebogens wurde an einer Stichprobe von überwiegend Studierenden (N = 752) in einer Onlinebefragung getestet. Die einfaktorielle Struktur der YFAS konnte bestätigt werden, bei einer zufriedenstellenden internen Konsistenz (Cronbach's α = .81). Konvergente Validität zeigte sich in mittleren bis hohen Korrelationen mit anderen Maßen problematischen Essverhaltens, während sich die YFAS hinsichtlich der diskriminanten Validität von anderen, aber dennoch verwandten Konstrukten, wie Alkoholabhängigkeit und Impulsivität, unterschied. Weiterhin zeigte sich die YFAS als signifikanter Prädiktor für die Häufigkeit von Essanfällen. Die YFAS scheint somit ein brauchbares Instrument für die Erfassung essensbezogener Verhaltensweisen darzustellen, die einen suchthaften Charakter aufweisen. [less ▲]

We introduce the concept of quasi-Lov\'asz extension as being a mapping $f\colon I^n\to\R$ defined over a nonempty real interval $I$ containing the origin, and which can be factorized as $f(x_1,\ldots,x_n)=L(\varphi(x_1),\ldots,\varphi(x_n))$, where $L$ is the Lov\'asz extension of a pseudo-Boolean function $\psi\colon\{0,1\}^n\to\R$ (i.e., the function $L\colon\R^n\to\R$ whose restriction to each simplex of the standard triangulation of $[0,1]^n$ is the unique affine function which agrees with $\psi$ at the vertices of this simplex) and $\varphi\colon I\to\R$ is a nondecreasing function vanishing at the origin. These functions appear naturally within the scope of decision making under uncertainty since they subsume overall preference functionals associated with discrete Choquet integrals whose variables are transformed by a given utility function. To axiomatize the class of quasi-Lov\'asz extensions, we propose generalizations of properties used to characterize the Lov\'asz extensions, including a comonotonic version of modularity and a natural relaxation of homogeneity. A variant of the latter property enables us to axiomatize also the class of symmetric quasi-Lov\'asz extensions, which are compositions of symmetric Lov\'asz extensions with $1$-place nondecreasing odd functions. Bidirectional transformations (bx) are a mechanism for maintaining the consistency of two (or perhaps more) related sources of information. Such sources can be databases, software models, documents, or their abstract models like graphs or trees. BX are an emerging topic in a wide range of research areas with prominent presence at top conferences in different fields. The methodologies used for bx range from classical program transformation to graph transformation techniques, from ad-hoc techniques for data synchronization to the development of domain-specific languages and their integration. The First International Workshop on Bidirectional Transformations (BX 2012) established a dedicated venue for bx in all relevant areas. It was initiated by the participants of the Dagstuhl Seminar "Bidirectional Transformations" (bx) in Germany 2011, which continued the GRACE International Meeting on Bidirectional Transformations held in Japan 2008. The workshop BX 2012 took place at the Tallinn University of Technology, Estonia on the 25th of March 2012, as a satellite event of ETAPS 2012, the European Joint Conferences on Theory and Practice of Software. The tool HENSHIN is an Eclipse plug-in supporting visual modeling and execution of rule-based EMF model transformations. This paper describes the recent extensions of HENSHIN by a visual editor for triple graph grammars (TGGs). The visual editor (called HENSHINTGG) supports a compact visualization of triple rules in an integrated editor panel. Internally, triple graph rules are represented as HENSHIN rules and can be simulated using the HENSHIN EMF model transformation engine. Our extension supports the automatic generation of forward translation rules for transforming source into target models. A converter from HENSHIN TGG rules to the graph transformation analysis tool AGG allows a systematic check for conflicts of forward translation rules in AGG based on critical pair analysis. [less ▲]

Here we present an ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) method for extracellular measurements of known and unexpected metabolites in parallel. The method was developed by testing 86 metabolites, including amino acids, organic acids, sugars, purines, pyrimidines, vitamins, and nucleosides, that can be resolved by combining chromatographic and m/z dimensions. Subsequently, a targeted quantitative method was developed for 80 metabolites. The presented method combines a UPLC approach using hydrophilic interaction liquid chromatography (HILIC) and MS detection achieved by a hybrid quadrupole-time of flight (Q-ToF) mass spectrometer. The optimal setup was achieved by evaluating reproducibility and repeatability of the analytical platforms using pooled quality control samples to minimize the drift in instrumental performance over time. Then, the method was validated by analyzing extracellular metabolites from acute lymphoblastic leukemia cell lines (MOLT-4 and CCRF-CEM) treated with direct (A-769662) and indirect (AICAR) AMP activated kinase (AMPK) activators, monitoring uptake and secretion of the targeted compound over time. This analysis pointed towards a perturbed purine and pyrimidine catabolism upon AICAR treatment. Our data suggest that the method presented can be used for qualitative and quantitative analysis of extracellular metabolites and it is suitable for routine applications such as in vitro drug screening. The E-pathway of transmembrane proton transfer has been demonstrated previously to be essential for catalysis by the diheme-containing quinol:fumarate reductase (QFR) of Wolinella succinogenes. Two constituents of this pathway, Glu-C180 and heme bp ring C (b(D)-C-) propionate, have been validated experimentally. Here, we identify further constituents of the E-pathway by analysis of molecular dynamics simulations. The redox state of heme groups has a crucial effect on the connectivity patterns of mobile internal water molecules that can transiently support proton transfer from the b(D)-C-propionate to Glu-C180. The short H-bonding paths formed in the reduced states can lead to high proton conduction rates and thus provide a plausible explanation for the required opening of the E-pathway in reduced QFR. We found evidence that the b(D)-C-propionate group is the previously postulated branching point connecting proton transfer to the E-pathway from the quinol-oxidation site via interactions with the heme bp ligand His-C44. An essential functional role of His-C44 is supported experimentally by site-directed mutagenesis resulting in its replacement with Glu. Although the H44E variant enzyme retains both heme groups, it is unable to catalyze quinol oxidation. All results obtained are relevant to the QFR enzymes from the human pathogens Campylobacter jejuni and Helicobacter pylori. [less ▲]
