Signal transduction through cytokine receptors is mediated mainly by non-covalently associated Jak tyrosine kinases. By confocal microscopy, the cytokine receptor gp130 and Jak1, fused with either yellow (YFP) or cyan (CFP) fluorescent protein, were found to be colocalized predominantly at intracellular vesicular structures and at the plasma membrane. Quantitative fluorescence recovery after photobleaching (FRAP) analysis at the plasma membrane revealed equal mobilities for gp130-YFP and Jak1-YFP. Thus, Jak1-YFP diffuses like a transmembrane protein indicating that membrane-bound Jak1 does not exchange rapidly with cytosolic Jaks. Applying a novel dual-color FRAP approach we found that immobilization of gp130-CFP by a pair of monoclonal antibodies led to a corresponding immobilization of co-transfected Jak1-YFP. We conclude from these findings that Jak1, once bound to a gp130 molecule, does not exchange between different receptors at the plasma membrane neither via the cytoplasmic compartment nor via a membrane-associated state. Even though the important role of early institutional language teaching has been proved by numerous studies, the practical applications of these findings are still very rare. In the Saarland, one of the German Laender which is situated on the border with France, a pilote project examinates the learning behaviour and mechanism with German primary school children learning French, their neighbouring language, from age 6 on. The author is the coordinator of the scientific team which follows the project from a universitarian point of view. The aim of the paper is to show the important role code-switching plays in this kind of teaching environment and it tries to evaluate its importance by giving examples from different stages of progress during the first two years of learning. It was the aim of this study to further explore the functional role of vitamin D in the endocrine pancreas. By gene targeting, we have recently generated mice in which a lacZ reporter gene is driven by the endogenous vitamin D receptor (VDR) promoter. These mice express a functionally inactive mutant VDR. Pancreatic islets but not exocrine pancreas cells showed strong lacZ reporter gene expression in mutant mice. To rule out possible influences of hypocalcemia on pancreatic endocrine function, a rescue diet enriched with calcium, phosphorus, and lactose was fed to wild-type (WT) and VDR mutant mice. The rescue diet normalized body weight and mineral homeostasis in VDR mutants. In glucose tolerance tests, baseline blood glucose levels were unchanged in fasting VDR mutants. However, blood glucose was elevated after oral or subcutaneous glucose loading, and maximum serum insulin levels were reduced by approximately 60% in VDR mutants vs. WT mice on either diet. In addition, insulin mRNA levels were decreased in VDR mutant mice on both diets, whereas pancreatic beta cell mass, islet architecture, and islet neogenesis were normal. These findings clearly establish a molecular role of the vitamin D-responsive elements in pancreatic insulin synthesis and secretion in vivo. [less ▲]

The peptide transporter PEPT2 mediates the cellular uptake of di- and tripeptides and selected drugs by proton-substrate cotransport across the plasma membrane. PEPT2 was functionally identified initially in the apical membrane of renal tubular cells but was later shown to be expressed in other tissues also. To investigate the physiological importance of PEPT2 and for a detailed analysis of the protein expression sites, we generated a Pept2 knockout mouse line in which the Pept2 gene was disrupted by insertion of a beta-galactosidase gene under the control of the PEPT2 promoter. The Pept2(-/-) mice showed no obvious phenotypic abnormalities but also no adaptive upregulation in the expression level of related genes in the kidney. The importance of PEPT2 in the reabsorption of filtered dipeptides was demonstrated in knockout animals by significantly reduced renal accumulation of a fluorophore-labeled and a radiolabeled dipeptide after in vivo administration of the tracers. This indicates that PEPT2 is the main system responsible for tubular reabsorption of peptide-bound amino acids, although this does not lead to major changes in renal excretion of protein or free amino acids. Dynamic processes such as cell migration and division depend on the actin cytoskeleton, a dense meshwork of protein polymers capable of undergoing rapid cycles of assembly and disassembly, under the control of a large number of actin-associated proteins. In cancer cells, structural and functional perturbations of the actin cytoskeleton correlate with higher proliferation rates and uncontrolled movement. Therefore, small molecules that act on the actin cytoskeleton of tumour cells and thus inhibit cell division and movement, may be of high therapeutic value. The dynamic properties of the actin cytoskeleton and the mechanism of action of actin-targeting drugs will be described. [less ▲]

Cette contribution propose d’explorer les processus de dispersion des ménages et de ségrégation par le revenu au sein d’un espace périurbain. La méthode consiste à simuler un modèle économique urbain avec deux types d’externalités de voisinage sur un espace cellulaire. Il permet la mise en place de processus dynamiques locaux endogènes et offre un éclairage sur la relation qui unit la morphologie des espaces périurbains et les caractéristiques des agents économiques qui s’y localisent. Des structures spatiales diverses sont générées sur deux configurations données des lots fonciers. Les équilibres présentent des structures concentriques constituées de multiples anneaux mais aussi, au cours des étapes de la croissance de la ville, des processus de dégradation cumulée ou des structures plus indépendantes de la distance.
